摘要
通过超声辅助Osborne分级法对新型蛋白质源乙醇梭菌蛋白进行分级提取,得到水溶性蛋白、盐溶性蛋白、醇溶性蛋白和碱溶性蛋白4种蛋白质,蛋白含量的大小顺序为碱溶性蛋白>水溶性蛋白>盐溶性蛋白>醇溶性蛋白,对其中含量最多的碱溶性蛋白的提取条件进行优化,结果表明,碱溶性蛋白的最佳提取条件为提取温度50℃、料液比1∶35(g/mL)、提取时间30 min、NaOH溶液质量分数0.3%。此条件下蛋白得率为75.75%。
Ultrasound-assisted Osborne fractionation was employed to extract proteins from Clostridium autoethanogenum.A total of 4 kinds of proteins were yielded:water-soluble protein,salt-soluble protein,alcohol-soluble protein and alkali-soluble protein.In terms of content,the 4 types of protein were in the order of alkali-soluble protein>water-soluble protein>salt-soluble protein>alcohol-soluble protein.Then the extraction conditions of the most abundant alkali-soluble protein were optimized as follows:extraction temperature of 50℃,material-to-liquid ratio of 1∶35(g/mL),extraction for 30 min,and NaOH solution at mass fraction of 0.3%.Under this condition,the yield of the protein was up to 75.75%.
