摘要
目的 探讨重组人干扰素-α1b联合布地奈德雾化对呼吸道合胞病毒(RSV)感染大鼠血清、肺泡灌洗液白介素-4(IL-4)的影响。方法 32只Wistar大鼠随机分为空白对照组、阳性对照组、重组人干扰素α-1b联合布地奈德雾化组(干扰素联合布地奈德组)、布地奈德雾化组,每组8只。除空白对照组外,其他3组给予7%水合氯醛灌肠及七氟烷麻醉后,均用RSV long株病毒(0.1 ml/只)予大鼠滴鼻的方法建造RSV病毒感染模型,空白对照组给予等量0.9%氯化钠溶液(0.1 ml/只)滴鼻。从实验第4天开始,空白对照组、阳性对照组分别给予0.9%氯化钠溶液(10 g/kg)雾化干预,干扰素联合布地奈德组予重组人干扰素-α1b注射液(15μg/次)联合吸入用布地奈德混悬液(0.5 mg/次)雾化干预,布地奈德组单纯予吸入用布地奈德混悬液(0.5 mg/次)雾化干预,均2次/d,连续雾化5 d。采用双抗体夹心ELISA法检测4组大鼠血清、肺泡灌洗液中IL-4的浓度。结果 与空白对照比较,阳性对照组血清、肺泡灌洗液中IL-4水平含量均升高(P<0.05);与阳性对照组比较,干扰素联合布地奈德雾化组、布地奈德雾化组血清、肺泡灌洗液中IL-4水平含量均偏低(P<0.05);与布地奈德雾化组比较,干扰素联合布地奈德雾化组血清、肺泡灌洗液中IL-4含量降低(P<0.05)。结论 重组人干扰素α-1b联合布地奈德雾化对RSV感染模型大鼠均有确切的治疗作用,可有效降低RSV感染大鼠血清和肺泡灌洗液中IL-4水平,进而抑制炎性反应,发挥积极治疗作用。
Objective To investigate the effects of recombinant human interferon-α1B combined with budesonide atomization on the levels of interleukin-4(IL-4)in serum and alveolar lavage fluid of rats infected with respiratory syncytial virus(RSV).Methods A total of 32 Wistar rats were randomly divided into blank control group,positive control group,recombinant human interferonα1b combined with budesonide atomization group and budesonide atomization group,with 8 rats in each group.Except for the rats in blank control group,the rats in the other three groups were given 7%chloral hydrate enema and sevoflurane anesthesia,and the rats were given RSV long strain virus(0.1ml/mouse)nasal drops to establish the virus infection models.The rats in blank control group were given the same amount of 0.9%sodium chloride solution(0.1ml/mouse)by nasal drops.From the fourth day of the experiment,the rats in blank control group and positive control group were given normal saline(10g/kg)atomization,and the rats in recombinant human interferonα-1b combined with budesonide(0.5mg/time)atomization were given recombinant human interferonα-1b(15μg/time)combined with budesonide(0.5mg/time)atomization,and the rats in budesonide group were treated by budesonide atomization(0.5mg/time),twice a day for 5 days.The levels of IL-4 in serum and alveolar lavage fluid were detected by double-antibody sandwich ELISA.Results Compared with those in blank control group,the levels of IL-4 in serum and alveolar lavage fluid in positive control group were significantly increased(P<0.05).Compared with those in positive control group,the levels of IL-4 in serum and alveolar lavage fluid in rIFNα-1B combined with budesonide atomization group and budesonide atomization group were significantly decreased(P<0.05).Conclusion The recombinant human interferon-α1B combined with budesonide atomization has definite therapeutic effects for the rats infected by RSV,which can obviously decrease the the levels of IL-4 in serum and alveolar lavage fluid of rats,so as to inhibit inflammatory response and play a positive therapeutic role.
