羟考酮对HepG2和SMMC-7721人肝癌细胞生长影响的体外研究

摘要 目的探讨羟考酮对HepG2和SMMC–7721人肝癌细胞增殖抑制的可能机制。方法体外培养HepG2和SMMC–7721人肝癌细胞,用CCK–8法检测不同浓度羟考酮对HepG2和SMMC–7721人肝癌细胞增殖的影响并计算药物IC50。细胞划痕实验检测不同浓度羟考酮对HepG2和SMMC–7721人肝癌细胞迁移的影响。Transwell实验检测不同浓度羟考酮对HepG2和SMMC–7721人肝癌细胞侵袭的影响。结果CCK–8实验提示羟考酮对HepG2和SMMC–7721人肝癌细胞增殖有抑制作用,其中羟考酮抑制HepG2人肝癌细胞的IC50为1.236mg/ml,抑制SMMC–7721人肝癌细胞的IC50为1.461mg/ml,对两种肝癌细胞系的增殖抑制随着剂量增大而增强。细胞划痕实验和Transwell实验显示随着浓度的升高,羟考酮对HepG2和SMMC–7721人肝癌细胞的迁移和侵袭有显著抑制作用,形态观察和细胞计数显示羟考酮可减少肝癌细胞的密度和数量,显著抑制肝癌细胞的集落形成并促进肝癌细胞凋亡。结论羟考酮具有抑制HepG2和SMMC–7721人肝癌细胞增殖、迁移和侵袭的作用。 Objective To investigate the possible mechanism of oxycodone inhibiting proliferation of HepG2 and SMMC–7721 human hepatoma cells.Methods CCK–8 assay was used to detect the effects of oxycodone on prolife-ration of HepG2 and SMMC–7721 cells and calculate the drug IC50.The effects of different concentrations of oxycodone on the migration of HepG2 and SMM–C7721 cells were detected by cell scratch assay.The effects of dif-ferent concentrations of oxycodone on the invasion of HepG2 and SMM–C7721 cells were detected by Transwell as-say.Results Oxycodone had an inhibitory effect on the growth of HepG2 and SMMC–7721 cells.The IC50 of oxy-codone for inhibition of HepG2 cells was 1.236 mg/ml,and the IC50 of SMMC–7721 cells was 1.461 mg/ml.The in-hibition of the growth of two hepatoma cell lines was dose-dependent.The cell scratch assay and Transwell assay showed that oxycodone significantly inhibited the migration and invasion of HepG2 and SMMC–7721 cells with the increase of concentration.Morphological observation and cell count showed that oxycodone could reduce the density and number of hepatoma cells,significantly inhibit the formation of hepatoma cells colony and promote apoptosis of hepatoma cells.Conclusion Oxycodone can inhibit the proliferation,migration and invasion of HepG2 and SMMC–7721 human hepatoma cells.
