壮腰通络方对腰椎间盘退行性病变大鼠病理变化及血清炎症因子表达的影响 被引量:4

Effects of Zhuangyao Tongluo Prescription on Pathological Changes and Expression of Serum Inflammatory Factors in Rats with Lumbar Intervertebral Disc Degeneration

摘要 目的观察壮腰通络方对腰椎间盘退行性病变大鼠病理变化及血清炎症因子表达的影响,探讨其可能的作用机制。方法65只雄性SD大鼠随机分为空白组10只和造模组55只,采用手术损伤致失稳退变的方法制备腰椎间盘退行性病变大鼠模型,将成模大鼠分为模型组、腰痹通胶囊组及壮腰通络方高、中、低剂量组,给药组分别予相应药物灌胃,空白组和模型组予等体积生理盐水灌胃,1次/d,连续8周。Masson染色观察大鼠椎间盘组织病理变化,ELISA检测血清肿瘤坏死因子(TNF)-α、白细胞介素(IL)-6含量,全自动生化分析仪检测丙氨酸氨基转移酶(ALT)、天冬氨酸氨基转移酶(AST)、血尿素氮(BUN)、血肌酐(SCr)水平,免疫组化检测大鼠椎间盘组织聚集蛋白聚糖(Aggrecan)、基质金属蛋白酶(MMP)13表达。结果与空白组比较,模型组大鼠椎间盘组织髓核细胞明显减少,血清TNF-α、IL-6含量增加(P<0.05),椎间盘组织Aggrecan表达降低,MMP13表达升高(P<0.05);与模型组比较,各给药组大鼠椎间盘组织髓核细胞增加、排列较规则,血清TNF-α含量减少(P<0.05),壮腰通络方中剂量组和腰痹通胶囊组大鼠血清IL-6含量减少(P<0.05),壮腰通络方中、低剂量组和腰痹通胶囊组大鼠椎间盘组织Aggrecan表达升高,MMP13表达降低(P<0.05)。各组大鼠血清ALT、AST、BUN和SCr水平差异无统计学意义(P>0.05)。结论壮腰通络方可有效延缓大鼠腰椎间盘退行性病变,其作用机制可能与上调Aggrecan表达、降低MMP13及血清炎症因子水平有关。 Objective To observe the effects of Zhuangyao Tongluo Prescription on pathological changes and expression of serum inflammatory factors in rats with lumbar intervertebral disc degeneration;To discuss its possible mechanism of action.Methods Totally 65 male SD rats were randomly divided into blank group(10 rats)and modeling group(55 rats),and the rat model was prepared by the method of surgical injury leading to destabilization and degeneration.The model rats were randomly divided into model group,Yaobitong Capsule group and Zhuangyao Tongluo Prescription high-,medium-,low-dosage group.Each administration group was given relevant medicine for gavage.The blank group and the model group were given equal volume normal saline for gavage,once a day for 8 weeks.Masson staining was used to observe the histomorphological changes of intervertebral discs tissue.ELISA was used to detect the contents of TNF-αand IL-6 in serum.The levels of alanine aminotransferase(ALT),aspartate aminotransferase(AST),blood urea nitrogen(BUN)and serum creatinine(SCr)were detected by automatic biochemical analyzer.Immunohistochemical staining was used to observe the expression of Aggrecan and MMP13 in intervertebral discs tissue.Results Compared with the blank group,the number of nucleus pulposus cells in intervertebral disc tissue of the model group reduced significantly,the contents of TNF-αand IL-6 in serum increased significantly(P<0.05),the expression of Aggrecan in intervertebral discs tissue decreased significantly,and the expression of MMP13 increased significantly(P<0.05).Compared with the model group,the number of nucleus pulposus cells in intervertebral disc tissue in each administered group increased and arranged regularly,the content of TNFαin serum reduced significantly(P<0.05),the content of IL-6 in serum of rats in Zhuangyao Tongluo Prescription medium-dosage group and Yaobitong Capsule group reduced significantly(P<0.05),the expression of Aggrecan in intervertebral disc tissue of rats in Zhuangyao Tongluo Prescription medium-and low-dosage groups,and Yaobitong Capsule group increased significantly,and the expression of MMP13 was significantly decreased(P<0.05).There was no statistical significance in serum ALT,AST,BUN and SCr in each group(P>0.05).Conclusion Zhuangyao Tongluo Prescription can effectively delay the lumbar intervertebral disc degeneration of rats,and the effects may be related to promoting the expression of Aggrecan and reducing the levels of MMP13 and serum inflammatory factors.
