不同剂量朱砂对睡眠剥夺前后大鼠的GABA和血汞的影响

Effects of Different Doses of Cinnabar on GABA and Blood Mercury in Rats before and after Sleep Deprivation

摘要 目的:观察不同剂量朱砂对于睡眠剥夺大鼠脑脊液和血液中γ-氨基丁酸(GABA)的调节作用及外周血汞的代谢曲线。方法:通过睡眠剥夺造模成功后分别给予高、中、低剂量朱砂治疗。实验过程中观察睡眠剥夺前后大鼠的一般情况及治疗前后大鼠脑脊液和血液中GABA的含量;通过热分解齐化原子吸收光度法测定连续7 d血汞曲线。结果:与模型组比较,观察组能够明显改善睡眠剥夺大鼠的体质量、精神状态和皮毛状况(P<0.01);与模型组比较,观察组大鼠血液中GABA含量未见明显变化(P>0.05);与模型组比较,观察组大鼠脑脊液中GABA的含量显著减少(P<0.01);观察组在连续服用不同剂量朱砂5 d内血汞浓度总体呈上升趋势,以高剂量组最明显,低剂量组次之,中剂量组最慢。高剂量组的大鼠血汞含量较中、低剂量组显著升高(P<0.01);结论:朱砂能够改善睡眠剥夺症状可能通过γ-氨基丁酸来实现,但可能不是朱砂主要作用靶点,连续服用时血汞含量的用药安全需要进一步研究。 Objective:To observe the regulating effects of different doses of cinnabar onγ-aminobutyric acid(GABA)in cerebrospinal fluid and blood and the metabolic curve of mercury in peripheral blood of sleep-deprived rats.Methods:High,medium and low doses of cinnabar were given after sleep deprivation modeling.During the experiment,the general situation of rats before and after sleep deprivation and the content of GABA in cerebrospinal fluid and blood before and after treatment were observed.The blood mercury curve for 7 days was determined by thermal decomposition and atomic absorption spectrophotometry.Results:Compared with model group,body weight,mental state and fur condition of sleep deprived rats were improved significantly in treatment group(P<0.01).Compared with model group,there was no significant change in blood GABA content in treatment group(P>0.05).Compared with model group,the content of GABA in cerebrospinal fluid in treatment group was significantly decreased(P<0.01).In the treatment group,the blood mercury concentration showed an increasing trend within 5 days of continuous administration of different doses of cinnabar,the highest in the high-dose group,followed by the low-dose group and the lowest in the medium-dose group.The blood mercury content in high dose group was significantly higher than that in medium and low dose groups(P<0.01).Conclusion:Cinnabar can improve sleep deprivation symptoms through GABA,but it may not be the main target of cinnabar,and the safety of blood mercury content during continuous administration needs further study.
