摘要 [目的]研究不同种类消毒剂处理组合对半夏不同外植体污染率、成活率的影响,筛选出半夏适合进行组织培养的外植体种类以及相应消毒方式。[方法]分别以野生半夏、栽培半夏珠芽和块茎2个典型取材部位为外植体,设置不同消毒处理组合,进行污染率、成活率比较试验。[结果]野生半夏相同类型的外植体较栽培半夏污染率低、成活率高。野生、栽培半夏株芽作为外植体较块茎污染率低、成活率高。隶属函数值分析表明,半夏外植体最佳消毒方案为75%乙醇50 s、0.1%HgCl_(2)10 min,此时,野生半夏块茎、株芽的污染率分别为15%、10%,成活率均为73.89%,栽培半夏的块茎、珠芽的污染率分别为17.22%、11.11%,成活率分别为68.89%、70.56%。[结论]野生半夏株芽更适合作为半夏无菌苗培养的外植体,最佳消毒方案为75%乙醇50 s、0.1%HgCl_(2)10 min。 [Objective]To study the effects of different types of disinfectant treatment combinations on the contamination rate and survival rate of different explants of Pinellia ternata,and screen out the types of explants suitable for tissue culture of Pinellia ternata and the corresponding disinfection methods.[Method]Wild Pinellia,cultivated Pinellia ternata buds and tubers were used as explants,and different disinfection treatment combinations were set up to compare the contamination rate and survival rate.[Result]The same type of explants of wild Pinellia ternata had lower contamination rate and higher survival rate than cultivated Pinellia ternata.Wild and cultivated Pinellia ternata buds as explants had lower contamination rates and higher survival rates than tubers.The membership function value analysis showed that the optimal disinfection scheme of Pinellia ternata explants was 75%ethanol for 50 s,0.1% HgCl_(2) for 10 min.At this time,the contamination rates of wild Pinellia ternata tubers and plant buds were lower,15%and 10%,respectively.The survival rates were both 73.89%.The contamination rates of tubers and bulb buds of cultivated Pinellia ternata were 17.22%and 11.11%respectively,and the survival rates were 68.89%and 70.56%,respectively.[Conclusion]The buds of wild Pinellia ternata are more suitable as explants for aseptic seedling cultivation of Pinellia ternata.The optimal disinfection plan is 75%ethanol 50 s,0.1% HgCl_(2)10 min.
