摘要
目的基于抗氧化药效成分群测定瓜蒌提取物中总黄酮的含量。方法以芦丁、木犀草素、芹菜素、异槲皮苷、山奈酚5种单体化合物按1∶2∶1∶2∶3(质量比)混合制备瓜蒌提取物的抗氧化药效成分群。采用紫外-可见分光光度法,以抗氧化药效成分群建立标准曲线并测定瓜蒌提取物中总黄酮的含量。结果抗氧化药效成分群线性方程为:A=0.2324C-0.0107,r=0.9995,抗氧化药效成分群在0.18~1.45μg/mL的范围内线性关系良好,方法学验证符合要求。基于抗氧化药效成分群测定瓜蒌提取物中总黄酮的含量为0.0022%(RSD=0.05%),与采用芦丁等单一对照品建立标准曲线测定瓜蒌提取物中总黄酮的含量结果差异有统计学意义(P<0.01)。结论基于抗氧化药效成分群测定瓜蒌提取物中总黄酮的含量,方法可靠、稳定,更能真实反映瓜蒌提取物中总黄酮的含量。
Objective To determine the content of total flavonoids in the extract of Trichosanthes kirilowii Maxim based on the antioxidant active components.Methods The antioxidant active components of the extract was prepared by mixing the five monomer compounds of rutin,luteolin,apigenin,isoquercetin and kaempferol at the mass ratio of 1∶2∶1∶2∶3.The standard curve was established by Ultraviolet-visible spectrophotometry,with antioxidant active components and the content of total flavonoids in the extract determined.Results The linear equation of antioxidant active components was A=0.2324C-0.0107,r=0.9995,the linear relationship was good in the range of 0.18~1.45μg/mL,and the methodological verification met the requirements.The content of total flavonoids in the extract determined based on antioxidant active components was 0.0022%(RSD=0.05%),which was different from the content of total flavonoids in the extract of Trichosanthes kirilowii Maxim determined by standard curve established by using rutin and other single reference substances.And such difference is statistically significant(P<0.01).Conclusion The method for determining the content of total flavonoids in the extract of Trichosanthes kirilowii Maxim based on antioxidant active components is reliable and stable,and can more truly reflect the content of total flavonoids in this extract.
