耐寒美国红枫‘秋火焰’离体培养技术研究

摘要 在北方寒冷地区的绿化公司建立耐寒美国红枫的快繁体系,以增加北方寒冷地区尤其是东北地区的彩叶树种的种类。该试验以美国红枫‘秋火焰’(Acer rubrum‘Autumn Blaze’)为研究对象,以腋芽茎段为外植体进行离体培养,对外植体消毒方式、启动培养、继代增殖培养、壮苗培养,以及生根培养的培养基配方进行正交试验以及双因素方差分析。通过初代培养、继代增殖培养、壮苗培养和生根培养4个阶段建立快繁体系。结果表明,当消毒组合为5%NaClO+0.1%HgCl_(2)时,平均污染率为33.15%,平均存活率为60.98%,死亡率为5.87%;最佳增殖培养基为改良MS+0.4 mg/L CPPU+0.1 mg/L IBA+1.0 mg/L 6-BA,增殖系数为6.45;最佳壮苗培养基为改良MS+1.0 mg/L ZT+0.5 mg/L IBA,平均株高为6.15 cm,茎粗为21.41 mm;最佳生根诱导培养基为1/2 MS+0.1 mg/L NAA+0.1 mg/L IBA,生根率达100%。此快繁体系效率高并适合应用于生产。 In order to increase the types of color-leaf tree species in the cold northern regions,the greening company in the cold northern regions established a rapid propagation system of cold-tolerant American red maple,and increased the types of color-leaf tree species in the north,especially in the northeast.In this experiment,the‘Autumn Blaze’of Acer rubrum L was taken as the research object,and the axillary bud stem segment was used as the explant in vitro culture.The medium formula for the sterilization of the explant,start culture,subgeneration culture,strong seedling culture,and rooting culture was carried out by orthogonal experiment and two-way analysis of variance.The rapid propagation system was established through four stages of primary cultivation,secondary propagation cultivation,strong seedling cultivation,and rooting cultivation.The results showed that the disinfection combination was 5%NaClO+0.1%HgCl_(2),the average pollution rate was 33.15%,the average survival rate was 60.98%,and the mortality rate was 5.87%.The best proliferation medium was modified MS+0.4 mg/L CPPU+0.1 mg/L IBA+1.0 mg/L 6-BA,the multiplication coefficient was 6.45.The best seedling medium was modified MS+1.0 mg/L ZT+0.5 mg/L IBA,the average plant height was 6.15 cm,and the stem thickness was 21.41 mm.The best rooting induction medium was 1/2 MS+0.1 mg/L NAA+0.1 mg/L IBA,and the rooting rate was 100%.This rapid propagation system is highly efficient and suitable for production.
