Th22及其分泌的细胞因子在变应性鼻炎患者外周血中的表达

Expressions of Th22 and its secreted cytokines in peripheral blood of patients with allergic rhinitis

目的:检测变应性鼻炎(AR)患者外周血Th22及其分泌的细胞因子的表达情况并分析其相关机制。方法:本研究共纳入30例AR患者和30例健康志愿者(HCs),ELISA检测AR患者及HCs外周静脉血中IL-22含量。流式细胞术检测AR患者及HCs外周血中Th22比例,将AR患者外周血中的PBMC平均分为3组,分别为空白对照组、同型对照组(加入PEMouse IgG2a,k)和实验刺激组(TNF-α处理组),并应用流式细胞术检测TNF-α对AR患者外周血中Th22的影响。结果:与HCs相比,AR患者外周血中CD4^(+)T细胞、IL-22表达水平显著提高(P<0.05)且AR患者外周血中IL-22表达水平与CD4^(+)T细胞中Th22呈正相关(r=0.966,P<0.001);HCs外周血IL-22表达水平与CD4^(+)T细胞中Th22比例呈正相关(r=0.981,P<0.001);AR患者外周血实验刺激组CD4^(+)T细胞中Th22比例明显高于空白对照组(P<0.05)。结论:AR患者外周血中IL-22表达水平升高,CD4^(+)T细胞中Th22比例提高,且Th22的比例与IL-22的表达水平呈正相关,AR患者外周血中,TNF-α可促进Th22的生成。

Objective:To detect expressions of Th22 and its secreted cytokines in peripheral blood of patients with allergic rhinitis(AR)and to analyze its related mechanisms.Methods:In this study,thirty AR patients and thirty healthy volunteers were(HCs)enrolled.ELISA was used to detect IL-22 levels in peripheral venous blood of AR patients and HCs.Flow cytometry was used to detect proportion of Th22 in peripheral blood of AR patients and HCs.PBMCs in peripheral blood of AR patients were divided into 3 groups:blank control group,isotype control group(PE-Mouse IgG2a,k)and experimental stimulation group(TNF-α treatment group),and applied flow cytometry to detect effect of TNF-α on Th22 in peripheral blood of AR patients.Results:Compared with HCs,expression levels of CD4^(+)T cells and IL-22 in peripheral blood of AR patients increased significantly(P<0.05),and expression levels of IL-22 in peripheral blood of AR patients were positively correlated with Th22 in CD4^(+)T cells(r=0.966,P<0.001);expression level of IL-22 in peripheral blood of HCs is positively correlated with proportion of Th22 in CD4^(+)T cells(r=0.981,P<0.001);proportion of Th22 in CD4^(+)T cells in peripheral blood experimental stimulation group of AR patients was significantly higher than that of blank control group(P<0.05).Conclusion:Expression level of IL-22 in peripheral blood of AR patients was increased,proportion of Th22 in CD4^(+)T cells was increased,and proportion of Th22 is positively correlated with expression level of IL-22.In peripheral blood of AR patients,TNF-α can promote Th22 generation.
