摘要
目的:探讨泌尿清丸对慢性非细菌性前列腺炎模型大鼠的影响及作用机制。方法:选取65只SD雄性大鼠,随机抽取10只作为空白组,剩余55只建立慢性非细菌性前列腺炎大鼠模型。造模成功后的大鼠随机分为模型组、阳性对照组及泌尿清丸低、中、高剂量组,每组10只。各组大鼠于第7 d开始灌胃给药,空白组和模型组予以蒸馏水;阳性对照组予以普乐安片,给药剂量为2 g/kg(相当于成人日用量的20倍),用生理盐水制备成0.2 g/ml的混悬液使用;泌尿清丸低、中、高剂量组予以泌尿清丸,给药剂量分别为4 g/kg、8 g/kg、16 g/kg(分别相当于成人日用量的5、10、20倍),用生理盐水分别制备成0.4 g/ml、0.8 g/ml、1.6 g/ml的混悬液使用。各组大鼠给药1次/日,灌胃容积均为10 ml/kg,连续45 d。给药结束后处死动物,称重前列腺质量,计算前列腺指数;制作各组大鼠前列腺组织切片染色,免疫组化检测大鼠前列腺组织中白细胞介素8(IL-8)、肿瘤坏死因子α(TNF-α)、白细胞介素1β(IL-1β)和白细胞介素4(IL-4)、白细胞介素6(IL-6)及白细胞介素10(IL-10)的表达。结果:与模型组比较,泌尿清丸低、中、高剂量组前列腺湿重及指数均明显减少(P<0.05),前列腺组织中的白细胞数目均明显减少,卵磷脂小体密度均明显升高(均P<0.05)。与模型组比较,泌尿清丸低、中、高剂量组均能明显降低前列腺组织中IL-8、TNF-α、IL-1β的水平,升高前列腺组织中IL-4、IL-6、IL-10的水平(均P<0.05)。结论:泌尿清丸治疗慢性非细菌性前列腺炎可能是通过降低炎症因子水平、升高抗炎因子水平而发挥作用。
Objective:To investigate the effect and mechanism of urinary clearing pill on chronic non-bacterial prostatitis model rats.Methods:Sixty-five SD male rats were selected into this study,ten SD male rats were randomly selected as blank group,and the remaining 55 rats were used to establish chronic non-bacterial prostatitis rat model.The rats were randomly divided into a model group after modeling,a positive control group and a low-dose,mediumdose and high-dose group of urinary clearing pill,with 10 rats in each group.The rats in each group were given intragastric administration on the 7 th day,and the blank group and model group were given distilled water;the positive control group was given Pule’an Tablets at a dose of 2 g/kg(equivalent to 20 times the adult dose for daily use),with Physiological saline was prepared as a 0.2 g/ml suspension;the low-dose,medium-dose and high-dose groups of were given urinary clearing pill,and the doses were 4 g/kg,8 g/kg,and 16 g/kg(equivalent to adult doses for daily use,respectively,5,10,and 20 times),prepare the suspensions of 0.4 g/ml,0.8 g/ml and 1.6 g/ml with physiological saline respectively.The rats in each group were administered once a day,and the gavage volume was 10 ml/kg for 45 consecutive days.After the administration,the animals were sacrificed,the weight of the prostate was weighed,and the prostate index was calculated;the prostate tissue sections of the rats in each group were made for staining,and the expression of interleukin-8(IL-8),tumor necrosis factorα(TNF-α),interleukin 1β(IL-1β),interleukin 4(IL-4),interleukin 6(IL-6)and interleukin 10(IL-10)were detected by immunohistochemistry.Results:Compared with the model group,the wet weight and index of the prostate in the low-dose,middle-dose and high-dose groups of urinary clearing pill were significantly decreased(P<0.05),the number of leukocytes in the prostate tissue was significantly decreased,and the density of lecithin body was significantly increased(both P<0.05).Compared with the model group,the low-dose,medium-dose and high-dose groups of urinary clearing pill can significantly reduce the levels of IL-8,TNF-αand IL-1βin prostate tissue,and increase the levels of IL-4,IL-6 and IL-10 in prostate tissue.(all P<0.05).Conclusion:Urinary clearing pill in the treatment of chronic non-bacterial prostatitis works by reducing the level of inflammatory factors and increasing the level of anti-inflammatory factors.
