摘要
目的:优化大花红景天总黄酮的提取工艺,提高大花红景天总黄酮提取效率。方法:采用超声辅助提取法提取大花红景天药材中有效成分,在单因素试验基础上,选取乙醇浓度、提取温度、提取时间3个因素进行Box-Behnken中心组合设计,以总黄酮提取率为指标,利用响应曲面法优化大花红景天总黄酮提取的最佳工艺。结果:大花红景天总黄酮最佳提取工艺条件为超声提取时间15min,乙醇浓度25%,超声提取温度40℃,在此条件下提取率为23.89%。结论:该模型回归性良好,试验拟合程度高、经验证该方法准确可靠,可用于大花红景天总黄酮的提取,为大花红景天资源化学的开发利用提供科学依据。
The extraction process of Rhodiola crenulata(HooK. f. et.Thoms) H. Ohba was optimized by response surface optimization ultrasonic assisted method, to provide scientific basis for optimizing the extraction of effective components from R.crenulata. Methods: On the basis of single factor experiment, the effects of ethanol concentration, ultrasonic temperature and ultrasonic time on the extraction yield amount of total flavonoids in R.crenulata were studied by response surface test. Results: The results showed that the optimal extraction parameters of total flavonoids were: Ethanol concentration of 25%,ultrasonic time was 15 min, ultrasonic temperature 40℃. Under this condition, the extraction rate is 23.89% Conclusion: The response surface optimization ultrasonic-assisted extraction method effectively improved the extraction yield amount of flavonoids from R.crenulata,the model has good regression and high fitting degree, to provide effective extraction method basis for the development and utilization of R.crenulata.
