基于网络药理学探究紫锥菊增强免疫功能的作用机制 被引量:1

Study on the mechanism of Echinacea purpurea in enhancing immunity based on network pharmacology

摘要 为了探究紫锥菊增强免疫功能的作用机制,试验基于网络药理学,利用NAPSS与Phytochem数据库收集紫锥菊活性成分,在NCBI数据库中收集免疫抑制的相关靶点,对活性成分与靶点取交集获得紫锥菊增强免疫功能的预测靶点。利用Cytoscape 3.7.2软件绘制紫锥菊活性成分-作用靶点网络图,利用String数据库构建蛋白质相互作用(PPI)网络。最后利用Metascape数据库,对紫锥菊增强免疫功能的生物过程进行GO富集分析,对其相关的信号通路进行KEGG信号通路分析。结果表明:共获得咖啡酸、阿魏酸、对香豆酸、原儿茶酸、槲皮素、芦丁等34个可增强机体免疫功能的紫锥菊活性成分,以及27个预测靶点;在PPI网络中,平均度值大于7.48的靶点有9个,分别为STAT3、TNF、CCND1、MTOR、IL-2、TLR4、RELA、TLR9、EZH2;GO富集分析涉及激酶活性的正调控、转移酶活性的正调控、对细菌来源分子的反应、对外界刺激反应的正调控、白细胞分化、凋亡信号通路、神经元死亡的调控等;KEGG信号通路分析涉及癌症中的蛋白多糖、P13K-Akt信号通路、Ras信号通路、调节干细胞多能性的信号通路、mTOR信号通路、癌症的中心碳代谢、AMPK信号通路、自噬的调控等信号通路。说明基于网络药理学预测紫锥菊活性成分可通过调节转录因子、免疫细胞、炎症因子等增强机体免疫功能。 In order to explore the mechanism of Echinacea purpurea in enhancing immunity based on network pharmacology,the active ingredients of Echinacea purpurea were collected from NAPSS and Phytochem platform,and the related targets of immunosuppression were collected from NCBI database.The intersection of drug and disease targets can be used to predict the immune function of Echinacea purpurea.Cytoscape 3.7.2 software was used to map the active ingredients-target network,and String database was used to construct the protein interaction(PPI)network.GO enrichment analysis was performed in Metascape database to predict the biological process of Echinacea purpurea enhancing immunity,and KEGG pathway analysis was performed to predict the related signaling pathways.The results showed that 34 active components of Echinacea purpurea that can enhance immune function and 27 action targets were obtained,including caffeic acid,ferulic acid,p-coumaricacid,protocatechuic acid,quercetin and rutin.In PPI network,there were 9 targets with an average degree value greater than 7.48,which were STAT3,TNF,CCND1,MTOR,IL-2,TLR4,RELA,TLR9 and EZH2.GO enrichment analysis involved positive regulation of kinase activity,positive regulation of transferase activity,positive regulation of bacteria-derived molecules,positive regulation of external stimulus response,leukocyte differentiation,apoptosis signaling pathway,neuron death and so on.KEGG pathway analysis involved proteoglycan in cancer,P13K-Akt signaling pathway,Ras signaling pathway,regulation of stem cell pluripotency signaling pathway,mTOR signaling pathway,central carbon metabolism of cancer,AMPK signaling pathway,autophagy regulation and other signaling pathways.It indicated that the prediction of the active ingredients of Echinacea purpurea based on network pharmacology could enhance immunity by regulating transcription factors,immune cells and inflammatory factors.
