响应面分析法优化藜麦花青素提取工艺

Optimization of extraction process of anthocyanins from Chenopodium quinoa Willd.by response surface methodology

摘要 为了优化藜麦花青素的提取工艺,本文以青白藜1号(白藜麦)、青藜2号(黑藜麦)和贡扎4号(红藜麦)为试验材料,在单因素试验基础上,以花青素提取量为响应值,以料液比、浸提温度和浸提时间为自变量进行响应面试验。结果表明:在70%酸化乙醇(磷酸为酸化剂,pH 3.0,V/V)溶剂条件下,白藜麦、黑藜麦和红藜麦花青素的最佳提取参数分别为料液比1:6.3、1:6.7、1:7.0 g/mL,浸提温度49.0、47.0、48.0℃,浸提时间90、100、85 min。在该条件下,3种粒色藜麦花青素实测提取量为49.06-52.27μg/g,与响应曲面拟合方程的预测值(49.47~52.34μg/g)基本一致,提取物具有典型的花青素红外特征。本研究可为藜麦花青素的提取与利用提供理论依据。 Qingbaili No.1(white Chenopodium quinoa Willd.),Qingli No.2(black Chenopodium quinoa Willd.)and Gongza No.4(red Chenopodium quinoa Willd.)are taken as experimental materials to optimize the extraction process of anthocyanins.Taking the extraction amount of anthocyanins as response value,response surface test is conducted with the ration of material to liquid,extraction temperature and extraction time as independent variables on the basis of single-factor experiment.The results show that taking 70%acidified ethanol(phosphoric acid as acidifier,pH 3.0,V/V)as the solvent,the optimum extraction parameters of anthocyanins from white,black and red Chenopodium quinoa Willd.are as follows:the ratio of material to liquid is 1:6.3,1:6.7 and 1:7.0 g/mL.The extraction temperature is 49.0,47.0 and 48.0 ℃.The extraction time is 90,100 and 85 min.The range of anthocyanins extracted from three colors of Chenopodium quinoa Willd.is 49.06~52.27μg/g under the conditions,which is basically consistent with the predicted value of the response surface fitting equation(49.47〜52.34μg/g).The extracts show typical infrared characteristics of anthocyanins.This study can provide a theoretical basis for the extraction and utilization of Chenopodium quinoa Willd.anthocyanins.
