摘要
目的采用模糊层次分析法优选复方土茯苓止痒喷雾的最佳提取工艺路线,优化原处方提取方法。方法以4-氨基吡啶致小鼠瘙痒为瘙痒模型,以小鼠舔体次数、小鼠血清组胺浓度和小鼠背部皮肤病理切片为指标,选取指标性成分和药效学指标作为准则层,方中指标性成分落新妇苷含量、丹皮酚含量、升麻素苷含量、小鼠舔体次数和血清组胺浓度作为方案层,采用模糊层次分析法(fuzzy analytic hierarchy process,FAHP)为指标赋权,得到各工艺路线加权综合得分,考察复方土茯苓止痒喷雾最佳提取工艺路线。结果方案层指标权重(ω)由高到低依次为舔体次数(ω=0.260)、组胺浓度(ω=0.235)、落新妇苷含量(ω=0.185)、丹皮酚含量(ω=0.160)、升麻素苷含量(ω=0.160)。得到评分高低顺序为工艺路线一>工艺路线二,故确定复方土茯苓止痒喷雾的最佳提取工艺路线为工艺路线一,即全方醇提。病理切片结果也证实了醇提优于水提。结论选取化学指标结合药效学指标,通过FAHP赋权,优选复方土茯苓止痒喷雾的提取工艺路线,为临床经验方制剂的工艺优选提供思路与方法。
Objective To optimize the extraction process of Bingling antipruritic spray by fuzzy analytic hierarchy process(FAHP)and optimize the extraction method of the original prescription.Methods Taking 4-aminopyridine-induced pruritus in mice as the pruritus model,taking the number of licks,the concentration of serum histamine and the pathological section of back skin as the indexes,the index components and pharmacodynamic indexes were selected as the criterion layer,the content of astilbin,paeonol,cimicillin glycoside,the number of licks and the concentration of serum histamine as the scheme layer,and the fuzzy analytic hierarchy process(FAHP)were used as the index,the weighted comprehensive scores of each process were obtained to investigate the best extraction process of Bingling antipruritic spray.Results The index weight(ω)of scheme level from high to low were:licking times(ω=0.260),histamine concentration(ω=0.235),astilbin content(ω=0.185),paeonol content(ω=0.160),cimicifugin glycoside content(ω=0.160).The scores from high to low were:route one>route two.Therefore,the best extraction route for the spray is the route one,which was reflux extraction with achohol.The results of pathological sections also confirmed that alcohol extraction was better than water extraction.Conclusion The selection of chemical parameters combined with pharmacodynamic indicators and the weight of FAHP were the best way to optimize the extraction process of Bingling antipruritic spray.
