基于网络药理学探讨大补元煎治疗阿尔茨海默病的机制

摘要 目的利用网络药理学方法探讨大补元煎治疗阿尔茨海默病(Alzheimer disease,AD)的活性成分及作用机制。方法利用TCMSP数据库筛选大补元煎的活性成分及作用靶点,利用GeneCards、OMIM、TTD、DrugBank数据库筛选AD相关靶点,取交集得到大补元煎治疗AD的潜在作用靶点。将交集靶点导入STRING数据库,获取蛋白相互作用(protein-protein interaction,PPI)关系,利用Cytoscape软件构建PPI网络及中药—成分—靶点—通路网络。借助Metascape平台进行基因本体(gene ontology,GO)功能注释和KEGG通路富集分析。结果筛选出活性成分175个,潜在靶点187个。大补元煎治疗AD的主要活性成分为槲皮素、山奈酶、β-谷甾醇、豆甾醇等,高效应靶点为APP、STAT3、PTGS2、PTGS1、AR等,生物学通路涉及癌症通路、神经活性配体—受体相互作用、cAMP信号通路、p53信号通路、5-羟色胺能突触等。结论大补元煎通过多成分、多靶点、多途径影响AD进程,从而发挥治疗作用。 Objective To explore the active ingredients and mechanism of Dabuyuan decoction in the treatment of Alzheimer disease(AD)by network pharmacology.Methods The TCMSP database was used to screen the active ingredients and targets of Dabuyuan decoction,and the Genecards,OMIM,TTD and DrugBank database were used to screen AD-related targets,and the potential targets of Dabuyuan Decoction in the treatment of AD were obtained,and the intersections were used to obtain potential targets for the treatment of AD by Dabuyuan decoction.The intersection targets were imported into the STRING database to obtain the PPI relationship,and the Cytoscape software was used to construct the PPI network and the herbs-ingredients-targets-pathways network.The Metascape platform was used for GO function annotation and KEGG pathway enrichment analysis.Results 175 active ingredients and 187 potential targets were screened out.The main active ingredients of Dabuyuan decoction in the treatment of AD were quercetin,kaempferol,β-sitosterol,stigmasterol,etc..The high-efficiency targets were APP,STAT3,PTGS2,PTGS1,AR,etc..The biological pathways involved pathways in cancer.Neuroactive ligand-receptor interaction,cAMP signaling pathway,p53 signaling pathway,serotonergic synapse,etc..Conclusion Dabuyuan decoction could affect the process of AD through multiple ingredients,targets and pathways,and exert a therapeutic effect.
