CpG-ODN作为免疫耐受诱导剂的研究进展

摘要 免疫球蛋白E(immunoglobulin E,IgE)介导的过敏性疾病可以通过过敏原特异性免疫疗法(allergen-specific immunotherapy,ASIT)治疗。研究人员使用佐剂和免疫调节剂增强ASIT效果并提高其安全性。CpG-寡脱氧核苷酸(CpG-oligodeoxynucleotide,CpG-ODN)具有免疫耐受促进特性,是治疗过敏性疾病的理想免疫调节剂之一。研究发现,CpG-ODN的剂量对于通过募集浆细胞样树突状细胞(plasmacytoid dendritic cells,pDCs)促进免疫调节至关重要。低剂量CpG-ODN会引起炎症反应,高剂量CpG-ODN会引发免疫耐受。CpG-ODN对IgE介导的过敏性疾病表现出预防和治疗潜力。对CpG-ODN的种类、CpG-ODN对免疫细胞的影响以及CpG-ODN作为ASIT免疫耐受诱导剂的作用机制等方面的研究进展进行综述,以期为相关研究提供参考。 Immunoglobulin E(IgE)-mediated allergic disorders can be treated with allergen-specific immunotherapy(ASIT).Adjuvants and immunomodulators were utilized by researchers to improve the efficacy and safety of ASIT.CpG-oligodeoxynucleotide(CpG-ODN)is one of the best immunomodulators for the treatment of allergic disorders due to its ability to promote immunological tolerance.The doses of CpG-ODN were found to be critical for promoting immune regulation through the recruitment of plasmacytoid dendritic cells(pDCs).Low doses of CpG-ODN elicited inflammatory responses while high doses of CpG-ODN triggered immunological tolerance.CpG-ODN has potential for the prevention and treatment of IgE-mediated allergic disorders.This paper reviews the research progress of the types of CpG-ODN,their effects on immune cells,and the mechanism of CpG-ODN as ASIT immunological tolerance inducer,in hoping to serve as references for related researches.
