Slc9a1基因在小鼠植入前胚胎发育过程中的作用研究

摘要 大量研究表明NHE1(Na^(+)/H^(+)exchanger 1)在肿瘤细胞增殖和凋亡过程中发挥重要作用,但其在植入前胚胎发育过程中作用尚未明确的报道。为探究Slc9a1(NHE1)基因在植入前胚胎发育过程中的作用,本研究利用RNA干扰(RNAi)技术构建小鼠Slc9a1基因敲低胚胎模型,重点研究了Slc9a1敲低对囊胚谱系分化、细胞增殖和细胞凋亡的影响。通过对谱系分化标志性分子Cdx2和Oct4的表达分析及p53和p21 mRNA水平的定量分析,结果发现Slc9a1通过影响囊胚的谱系分化、细胞周期和细胞凋亡参与早期胚胎发育。 A large number of studies have shown that NHE1 plays an important role in the proliferation and apoptosis of tumor cells.However,the function of Slc9a1(NHE1)during preimplantation embryonic development has not been clearly reported.In order to explore the potential role of Slc9a1 gene in preimplantation embryonic development,this study researched the effect of Slc9a1 knockdown on blastocyst lineage differentiation,cell proliferation and apoptosis by using RNA interference(RNAi)technology to construct a model of Slc9a1 gene knockdown in mouse embryos.Through the expression analysis of the marker protein Cdx2 and Oct4 of lineage differentiation and the quantitative analysis of p53 and p21 mRNA levels,it was found that Slc9a1 was involved in early embryo development by affecting blastocyst lineage differentiation,cell cycle and apoptosis.
