摘要
目的近年来,恶性肿瘤膀胱癌在我国发病率和死亡率有一定上升趋势,本文基于网络药理学方法,预测中草药豆蔻治疗膀胱癌的潜在作用机制。方法通过中药系统药理学数据库(TCMSP)筛选出豆蔻的有效活性成分,在Uniport数据库将其活性成分检索出相对应的基因靶点。从GeneGards数据库获取膀胱癌的靶点基因并筛选出疾病和药物交集基因靶点。利用DAVID数据库对豆蔻治疗膀胱癌的基因靶点进行可视化GO功能和KEGG通路富集分析。结果豆蔻治疗膀胱癌主要成分是乔松素、槲皮素、木犀草素、熊竹素等;关键作用靶点为IL6、TP53、MYC、CCND1、AKT1、HIF1A、CASP3、JUN、PTEN、VEGFA、ERBB2等;主要参与通路为膀胱癌通路、癌症通路、PI3K-Akt信号通路等多条通路。结论本研究利用网络药理学方法分析豆蔻治疗膀胱癌的潜在活性成分和作用机制,为后续中草药豆蔻治疗膀胱癌实验研究和临床治疗提供一定的理论参考。
Objective In recent years,the incidence rate and mortality rate of malignant tumor in bladder cancer in China are increasing.Based on network pharmacology,the potential mechanism of Chinese herbal medicine cardamom in treating bladder cancer is predicted.Methods The effective active components of Amomum cardamon were screened through TCMSP database,and the corresponding gene targets were retrieved from Uniport database.The target genes of bladder cancer were obtained from the GeneGards database and the target genes for disease and drug interaction were screened out.The DAVID database was used to visualize the gene targets for the treatment of bladder cancer,and GO function and KEGG pathway enrichment analysis were performed.Results The main components of Amomum cardamon in treating bladder cancer are Pinocembrin,Quercetin,Luteolin,Jaranol,and so on;The key targets are IL6,TP53,MYC,CCND1,AKT1,HIF1A,CASP3,JUN,PTEN,VEGFA,ERBB2,etc;The main pathways involved are bladder cancer pathway,cancer pathway,PI3K-Akt signaling pathway and many other pathways.Conclusion This study used network pharmacology to analyze the potential components and mechanisms of Amomum cardamon in treatment of bladder cancer,so as to provide a theoretical reference for subsequent experimental research and clinical treatment.
