摘要
通过邻硝基苯-β-D-吡喃半乳糖苷(ONPG)法测定自有9株乳酸菌菌株的β-半乳糖苷酶活力,从中选择酶活力最高的菌株,进行小鼠灌胃试验,28 d后测定小鼠小肠β-半乳糖苷酶活性,进一步验证其对乳糖不耐症的益生作用。结果表明:动物双歧杆菌乳亚种(Bifidobacterium animails subsp.lactis)HCS04-002的β-半乳糖苷酶酶活力最高,为0.758 U/mL;小鼠试验中,动物双歧杆菌乳亚种(Bifidobacterium animails subsp.lactis)HCS04-002菌粉中剂量组(酶活力0.768 U/mL)和高剂量组(酶活力0.751 U/mL)试验小鼠小肠的β-半乳糖苷酶活性明显高于对照组(灌胃纯水,酶活力0.118 U/mL)和模型组(灌胃全脂乳粉,酶活力0.304 U/mL),说明动物双歧杆乳亚种HCS04-002具有一定的缓解乳糖不耐受的作用,可进一步研究其在治疗乳糖不耐受产品中的应用。
The β-galactosylase enzyme activity of 9 lactic acid bacteria strains was detected by the method of o-nitrobenzene-β-D-galactosylpyran(ONPG),and then the strain with the highest enzyme activity was chosen,which was used to do oral perfusion to mice and the activity of β-galactoside enzyme in small intestine of mice was detected after 28 d of oral perfusion.The results was a further proof of its probiotic effect on lactose intolerance.The results showed that Bifidobacterium animails subsp.Lactis HCS04-002 had the highest activity of β-galactosylase,which was 0.758 U/mL.In oral perfusion to mice,Bifidobacterium animails subsp.Lactis HCS04-002 in the medium dose group(enzyme activity 0.768 U/mL)and the high dose group(enzyme activity 0.751 U/mL)were significantly higher than that in the control group(ig pure water,enzyme activity 0.118 U/mL)and the model group(ig whole milk powder,enzyme activity 0.304 U/mL).These results indicated that Bifidobacterium animalis subsp.Lactis HCS04-002 showed a certain function of alleviating lactose intolerance and its application in treating lactose intolerance products deserved further investigation.
