地霉菌对发酵药渣中残余林可霉素的生物降解 被引量:1

摘要 筛选、鉴定出能高效降解林可霉素的酵母菌,并对其降解机理进行初步探究。采用平板降解活性测试及酶联免疫吸附法进行筛选,采用形态学及26S rRNA序列分析等进行鉴定。采用构建酵母菌基因组文库的方法筛选获得降解基因,并用异源表达、纯化、活性测试方法验证基因功能。从林可霉素生产药厂的废水废渣中筛选到一株酵母菌S1,对林可霉素具极高降解活性。鉴定为半乳糖霉菌Galactomyces sp.。菌株S1中的环氧化物水解酶基因dgs1在降解过程中起着重要作用。该基因的异源表达产物具有较强的降解林可霉素的功能,降解能力可达33.95%。获得一株林可霉素高效降解酵母菌Galactomyces sp.S1,其活性物质为环氧化物水解酶Dgs1,为今后林可霉素高效降解工程菌的构建及林可霉素药渣环保处理奠定基础。 Screening and identifying the high-efficiency degradation of lincomycin yeast,its degradation mechanism was preliminarily explored.Yeast strains of high-efficiency degradation of lincomycin were screened by plate degradation activity test and ELISA activity detection.Strains were identified by phenotypic and genotypic characteristics.The degradation gene was screened by constructing yeast genome library,and the gene functions were verified by heterologous expression,purification and activity testing methods.A yeast strain S1 was screened from waste water and residue from lincomycin production pharmaceutical factory,which had extremely high degrading activity on lincomycin.The yeast strain S1 was classified to be Geotrichum sp.The epoxide hydrolase gene dgs1 in strain S1 was identified to be the main degradation factor.The heterologous expression product of the gene dgs1 had a strong function of degrading lincomycin.The degradation capability of Rm-Dgs1 could reach 33.95%.A yeast strain Geotrichum sp.S1 with high degradation activity against lincomycin was isolated and the virulence factor was hydrolase Dgs1.The research results could lay a foundation for the construction of high-efficiency lincomycin degrading engineering bacteria and environmental protection treatment of lincomycin residue.
