响应面法优化暗褐脉柄牛肝菌粗多糖提取工艺 被引量:4

Optimization of Extraction Process for Crude Polysaccharide from the Fruiting Bodies of Phlebopus portentosus Based on Response Surface Methodology

摘要 以暗褐脉柄牛肝菌子实体粗多糖提取率为指标,在单因素试验基础上,利用Box-Behnken Design中心组合试验设计,考察液料比、浸提温度、提取时间3个因素对暗褐脉柄牛肝菌子实体粗多糖提取率的影响,优化暗褐网柄牛肝菌子实体粗多糖的提取工艺。得到最佳提取工艺条件:液料比20︰1 mL/g,浸提温度78.72℃,提取时间77.76min。调整最佳浸提温度为79℃,最佳提取时间为78 min,在此条件下所得暗褐脉柄牛肝菌子实体粗多糖提取率为15.95%,与模型预测值15.99%偏差较小,表明响应面优化的模型参数准确可靠。 The Box-Behnken design response surface method was used to optimize the extraction process of crude polysaccharide from the fruiting bodies of Phlebopus portentosus with the extraction rate of crude polysaccharide as the index on the basis of single factor test of liquid-solid ratio,extraction temperature and extraction time.The optimal extraction condition of crude polysaccharide from the fruiting bodies of P.portentosus was obtained as follows:liquid-solid ratio 20︰1 mL/g,extraction temperature 78.72℃,and extraction time 77.76 min.When the extraction temperature was adjusted to 79℃,and the extraction time was adjusted to 78 min,the extraction rate of the crude polysaccharide was 15.95%,and there was little deviation from the predicted value of 15.99%,which indicated that the model parameters of the response surface optimization were accurate and reliable.
