摘要
建立风味发酵乳中三氯蔗糖的高效液相色谱测定方法。样品用乙腈-水(50∶50,V/V)溶液提取,过HLB固相萃取柱净化富集浓缩。使用Agela Innoval ODS-2色谱柱(250 mm×4.6 mm,5μm)色谱柱分离,以乙腈和水为流动相进行等度洗脱,示差检测器检测,外标法定量。结果表明,三氯蔗糖在0.0015~0.1000 g/L范围内有良好线性关系,相关系数在0.999以上,检出限为0.001 g/kg,定量限为0.003 g/kg。风味发酵乳样品在3个水平(0.003,0.015和0.300 g/kg)下做加标试验,其平均回收率在90.0%~110%之间,相对标准偏差低于5%(n=6)。该方法前处理便于广泛推广应用,结果准确,适用于风味发酵乳中三氯蔗糖的快速检测。
To establish a method for determination of sucralose in flavored fermented milk by high performance liquid chromatographic,samples were extracted with acetonitrile-water(50∶50,V/V)solution,purified and concentrated by HLB solid phase extraction column.Agela Innoval ODS-2 chromatographic column(250 mm×4.6 mm,5μm)was used for separation,and acetonitrile and water were used as mobile phase for isotropic elution,detected by differential detector,and quantified by external standard method.The results showed that sucralose had good linearity in the range of 0.0015-0.1000 g/L;The correlation coefficient was above 0.999;The detection limit was 0.001 g/kg,and the quantitative limit was 0.003 g/kg.For flavor fermented milk samples,the standard was added at 3 levels(0.003,0.015 and 0.300 g/kg),and the average recovery rate was 90.0%-110%;The relative standard deviation was less than 5%(n=6).The pre-treatment of the method is convenient for wide promotion and application,and the result is accurate.It is suitable for the rapid detection of sucralose in flavored fermented milk.
