摘要
目的初探生地黄归经的物质基础。方法将生地黄以10 g/kg生药剂量连续灌胃给予大鼠7 d,末次给药后15、30、60、90、120、180 min分别取血和心、肝、肾、脾、肺、小肠、脑等组织,采用高效液相色谱法测定生地黄梓醇在大鼠体内各组织中的浓度并计算药动学相关参数,初步探索生地黄归经的物质基础。结果在既定的考察时间点,生地黄给药后在大鼠的血清及观测组织均不同程度检测到了梓醇,在组织中分布由高到底低前三名依次为肝、肾、心,此外在脾、脑、小肠、肺组织也有一定分布。结论初步表明梓醇能够作为表征生地黄归经的一个主要物质基础。
Objective To initially explore the material basis of Rehmannia glutinosa(DH)meridian tropism.Methods The rats were continuously administered to rats at a dose of 10 g/kg for 7 days,and blood and blood samples were taken 15,30,60,90,120,and 180 minutes after the last administration.In tissues such as heart,liver,kidney,spleen,lung,small intestine,brain,etc.,the concentration of catalpol,the major medicinal compound of DH,was determined by high performance liquid chromatography in various tissues of rats,and the relevant parameters of pharmacokinetics were calculated so as to initially explore the material basis of DH’s meridian tropism.Results At the established inspection time point,catalpol was detected in the rat’s serum and observation tissues to varying degrees after the administration of Rehmannia glutinosa.The top three distributions in the tissues were liver,kidney,and heart from high to low.In addition,catalpol is also distributed in the spleen,brain,small intestine,and lung tissues.Conclusion This study preliminarily shows that catalpol can be used as a main material basis for characterizing the meridian tropisma of Rehmannia glutinosa.
