摘要
垂体促卵泡激素(FSH)是卵泡生长发育、颗粒细胞增殖及类固醇合成、精子生成等生殖生理活动必需的糖蛋白激素(GPH),在人类和动物辅助生殖中扮演着非常重要的角色。天然FSH的半衰期很短,单次使用的有效血药浓度远不足以维持哺乳动物的卵泡周期,并且天然FSH的来源越来越受限。因此,重组FSH(rFSH)药物的开发迫在眉睫,rFSH不仅有助于提高繁殖效率,还可减少频繁给药给人类医学临床患者带来的精神压力和给动物造成的应激。本文主要针对近几年国内外rFSH的研究,如FSH的糖基化修饰、新生儿可结晶片段(Fc)受体(FcRn)介导及聚乙二醇(PEG)化,以及其对雌性卵巢调控功能进行综述,旨在为人类与动物的辅助生殖提供参考。
Pituitary follicle-stimulating hormone(FSH) is a necessary glycoprotein hormone,(GPH) for follicular growth and development, granulosa cell proliferation, steroid synthesis, spermatogenesis and other reproductive physiological activities,and plays a very important role in assisted reproduction in humans and animals. However, the half-life of natural FSH is very short, the effective blood concentration of single use is far from enough to maintain the follicular cycle of mammals, and the source of natural FSH is more and more limited. Therefore, the development of recognition FSH(rFSH)drugs is imminent.RFSH can not only improve the reproductive efficiency, but also reduce the stress of human medical clinical patients and animals caused by frequent drug administration. This paper mainly focuses on the research of new rFSH at home and abroad in recent years, such as glycosylation modification, the neonatal fragment crystallizable receptor(FcRn)mediation and polyethylene glycol(PEG)fusion of FSH, and summarizes the regulation function of female ovary, aining to provide reference for assisted reproduction of human and animal.
