摘要
目的:探讨抗支糖浆对雾化烟草提取物环境哮喘模型小鼠气道炎症及Th17/Treg免疫平衡的影响。方法:将90只SPF级KM雌性小鼠随机分为9组,空白组(A组)、模型组(B组)、抗支糖浆低剂量组(C组)、抗支糖浆高剂量组(D组)、阳性药组(E组)、烟雾模型组(F组)、烟雾抗支糖浆低剂量组(G组)、烟雾抗支糖浆高剂量组(H组)、烟雾阳性药组(I组)。烟雾模型组、烟雾抗支糖浆低剂量组、烟雾抗支糖浆高剂量组、烟雾阳性药组用雾化烟草提取物溶液雾化,除空白组外,其余各组给予OVA及Al(OH)_(3)注射致敏。给药组于实验第21天开始进行药物干预。观察哮喘小鼠模型一般状态及行为学变化情况,检测小鼠BALF中辅助性T细胞17(Th17)、调节性T细胞(Treg)相关细胞因子IL-10、IL-17、IL-23水平,过碘酸雪夫染色(PAS)观察肺组织病理变化,统计BALF中白细胞的分类及计数。结果:空白组小鼠在OVA激发前呼吸平稳,雾化过程中逐渐出现点头、喘息、前肢搔鼻、毛色光泽减低等表现,且随雾化激发次数的增加而加重。模型组、烟雾模型组症状相似,烟雾模型组小鼠雾化烟草提取物后,表现较模型组小鼠更为明显,药物干预后,小鼠状态得到改善。各组IL-10、IL-17、IL-23含量均有不同程度改变。空白组未见明显肺组织病理改变,模型组、烟雾模型组病理改变最为明显,与模型组、烟雾模型组比较,其余6组的病理变化显著减轻。除空白组外,其余8组的白细胞总数、EOS、LYM、NEU各组所占百分比均显著增多(P<0.05)。结论:抗支糖浆可改善雾化烟草提取物环境哮喘模型小鼠的一般病理状态,改善哮喘症状;抗支糖浆可通过上调IL-10水平,下调IL-17、IL-23的表达,恢复Th17/Treg平衡,改善气道炎症,减少黏液高分泌,达到治疗哮喘的目的。
Objective:To investigate the effects of Kangzhi Syrup on airway inflammation and Th17/Treg immune balance in atomized tobacco extract environmental asthma model mice.Methods:90 SPF KM female mice were randomly divided into the blank group,the model group,the low-dose group of Kangzhi Syrup,the high-dose group of Kangzhi Syrup,the positive drug group,the smoke model group,the smoke plus low-dose Kangzhi Syrup group,the smoke plus high-dose Kangzhi Syrup group,and the smoke plus positive drug group.The smoke model group,the smoke plus low-dose Kangzhi Syrup group,the smoke plus high-dose Kangzhi Syrup group,and the smoke plus positive drug group were atomized with atomized tobacco extract solution.Except the blank group,the other groups were sensitized by OVA and Al(OH)_(3) injections.The administration groups began drug intervention on the 21st day of the experiment.The general state and behavioral changes of asthmatic mice model were observed,the levels of helper T cell 17(Th17)and regulatory T cell(Treg)related cytokines of IL-10,IL-17 and IL-23 in BALF were measured,the pathological changes of lung tissues were observed by Periodate Schiff staining(PAS),and the classification and the count of leukocytes in BALF were counted.Results:The mice in the blank group breathed steadily before OVA stimulation,and gradually nodded,gasped,scratched nose with forelimbs and decreased hair color and luster during atomization,which increased with the increase of atomization stimulation times.The symptoms of the model group and the smoke model group were similar.After atomizing tobacco extract,the performance of the smoke model group mice was more obvious than that of the model group mice.After drug interventions,the state of mice was improved.The contents of IL-10,IL-17 and IL-23 in each group changed in varying degrees.There were no significant pathological changes in the blank group,and the pathological changes in the model group and the smoke model group were most obvious.Compared with those in the model group and the smoke model group,the pathological changes in the other six groups were significantly reduced.Except the blank group,the total number of leukocytes,the percentage of EOS,LYM and NEU in the other eight groups increased significantly.Conclusion:Kangzhi Syrup can improve the general pathological state and asthma symptoms of atomized tobacco extract environmental asthma model mice.Kangzhi Syrup can treat asthma by up-regulating the level of IL-10,down-regulating the expression of IL-17 and IL-23,restoring Th17/Treg balance,improving airway inflammation and reducing mucus hypersecretion.
