外源性内脂素对促排卵SD大鼠子宫内膜容受性的影响

Effect of exogenous endoliposin on the endometrial tolerance of ovulatory SD rats

摘要 目的 研究外源性内脂素对不同周龄促排卵SD大鼠子宫内膜容受性的影响及机制。方法 选年轻、大龄大鼠各40只,分为年轻组、大龄组,将2组均分为干预组、非干预组2个亚组,非干预组做促排卵,干预组做促排卵联合外源性内脂素干预。妊娠后第4天、第7天每组处死10只,对比雌二醇(estradiol,E2)、孕酮(progesterone,P)水平。观察第4天子宫内膜变化、微血管密度(microvessel density,MAD)、胞饮突数量、血管内皮生长因子(vascular endothelial growth factor,VEGF)-内皮型一氧化氮合酶(endothelial nitric oxide synthase,eNOS)-一氧化氮(nitric oxide,NO)通路因子表达量,分析VEGF、eNOS相关性;统计第7天妊娠率、着床胚泡数。结果 各组妊娠第7天E2、P高于妊娠第4天(P<0.05)。大龄非干预组E2、P、子宫内膜厚度、胞饮突数量、MAD、妊娠率、着床胚泡数、VEGF、eNOS mRNA表达量低于年轻非干预组,高于大龄非干预组(P<0.05);VEGF与eNOS正相关(r=0.94,P<0.001)。结论外源性内脂素可改善促排卵SD大鼠子宫内膜容受性,可能与VEGF-eNOS-NO通路介导子宫内膜血管生成相关,对年轻鼠无影响。 Objective To study the effect and mechanism of exogenous lactone on endometrial receptivity in SD rats with different weeks of ovulation induction. Methods Select 40 young rats and 40 old rats, and divide them into young group and old group. The two groups are divided into two subgroups: intervention group and non-intervention group. The non-intervention group is used for ovulation induction, and the intervention group is used for ovulation induction combined with exogenous Sex visfatin intervention. On the 4th and 7th day after pregnancy, 10 animals were sacrificed in each group,and the levels of estradiol(E2) and progesterone(P) were compared. Observe the changes of endometrium, microvessel density(MAD), pinocytosis number, vascular endothelial growth factor(VEGF)-endothelial nitric oxide synthase(eNOS) on the 4th day-nitric oxide(NO) pathway factor expression, analysis of the correlation between VEGF and eNOS;statistics on the 7th day pregnancy rate and the number of implanted blastocysts. Results E2 and P on the 7th day of pregnancy in each group were higher than those on the 4th day of pregnancy(P<0.05). The older non-intervention group E2, P, endometrial thickness,pinocytosis number, MAD, pregnancy rate, implantation blastocyst number, VEGF, eNOS mRNA expression were lower than the young non-intervention group, higher than the older non-intervention group(P<0.05). VEGF is positively correlated with eNOS(r=0.94, P<0.001). Conclusion Exogenous visfatin can improve the endometrial receptivity of ovulation-stimulating SD rats, which may be related to the VEGF-eNOS-NO pathway mediated endometrial angiogenesis, and has no effect on young rats.
