摘要 目的:探讨黄芪总黄酮(TFA)对血管性痴呆(VD)模型大鼠学习记忆损伤的干预作用与机制。方法:双侧颈总动脉结扎构建大鼠血管性痴呆模型,分别给予TFA 25,50,100 mg/kg连续治疗2周。观察TFA对大鼠神经功能评分、学习记忆损伤、血清神经特异性烯醇化酶(NSE)活性、脑组织ROCK2表达和Tau蛋白磷酸化的影响。结果:50,100 mg/kg TFA可明显改善血管性痴呆大鼠神经功能评分和学习记忆损伤。此外,血管性痴呆大鼠海马组织中ROCK2表达和Tau Ser262位点磷酸化明显增高,但50,100 mg/kg TFA治疗组大鼠海马组织中ROCK2表达和Tau Ser262位点磷酸化明显降低。结论:TFA可明显改善血管性痴呆大鼠的学习记忆损伤,其机制可能与抑制ROCK2表达和下调Tau磷酸化有关。 Objective:To investigate the intervention effect of total flavonoids of Astragalus(TFA)on learning and memory impairment in vascular dementia(VD)model rats,and to explore its mechanism.Methods:The rat model of vascular dementia was established by bilateral common carotid artery ligation,and treated with TFA 25,50 and 100 mg/kg for 2 weeks.The effects of TFA on neurological function score,learning and memory impairment,serum NSE activity,ROCK2 expression in brain tissue and Tau protein phosphorylation in rats were observed.Results:50,100 mg/kg TFA could significantly improve neurological function score and learning and memory impairment in vascular dementia rats.In addition,the expression of ROCK2 and phosphorylation of Tau Ser262 in hippocampus of vascular dementia rats were significantly increased,but the expression of ROCK2 and phosphorylation of Tau Ser262 in hippocampus of 50,100 mg/kg TFA treatment groups were significantly decreased.Conclusion:TFA can significantly improve the learning and memory impairment in vascular dementia rats,which may be related to the inhibition of ROCK2 expression and the down-regulation of Tau phosphorylation.
