肺腺癌中CDK1的生物信息学分析 被引量:1

摘要 目的:分析基于生物信息学对CDK1在肺腺癌(LUAD)中的表达和临床意义。方法:通过GEPIA分析CDK1的表达,在UALCAN数据库中分析肺腺癌分期、淋巴结转移等临床特征与CDK1表达的关系,GEPIA分析CDK1表达与肺腺癌预后的关系,Oncomine数据库挖掘CDK1的共表达基因,WebGestalt对共表达基因进行G0和KEGG通路富集分析。String数据库整合基因蛋白互作网络(PPI),并进行Cytoscape可视化分析,CytoHubba挑选核心基因。Kaplan-Meier plotter和GEPIA数据库对筛选出的hub基因进行了更多的分析和验证。结果:在肺腺癌中,CDK1表达明显高出正常组织(P<0.01)。UALCAN数据库分析表明肺腺癌患者的分期、性别等临床特征与CDK1的表达没有显著相关性。GEPIA分析显示,与CDK1高表达的肺腺癌患者相比,CDK1低表达患者的总生存率增加(P<0.05)。Oncomine数据库筛选出CDK1的共表达基因有TOP2A、MCM4等18个基因。GO分析显示,CDK1共表达基因主要与染色体定位、染色体凝集、ATP酶活性相关KEGG分析显示,CDK1共表达基因参与p53信号通路等进程。借助String、Cytoscape、CytoHubba,获得CDK1共表达基因的PPI网络,继而挑选hub基因,按Degree大小排列顺序选择5个hub基因,分别是CDC6、MAD2L1、TOP2A、KIAA0101、MCM4。最后,证实5个hub基因和肺腺癌患者的预后密切相关。结论:在肺腺癌患者中CDK1高表达,且与肺腺癌患者的预后息息相关,为肺腺癌的治疗及诊断提供重要指导。 Objective:Based on bioinformatics,the expression and clinical significance of CDK1 in lung adenocarcinoma(LUAD)were analyzed.Methods:GEPIA was used to analyze the expression of CDK1,the relationship between lung adenocarcinoma staging,lymph node metastasis and other clinical features and the expression of CDK1 was analyzed in the UALCAN database.The relationship between CDK1 expression and the prognosis of lung adenocarcinoma was analyzed by GEPIA,and the co-expressed genes of CDK1 were discovered in the Oncomine database.WebGestalt conducts G0 and KEGG pathway enrichment analysis of co-expressed genes.The String database integrates the gene-protein interaction network(PPI),and launches the Cytoscape visual analysis,and CytoHubba selects the core genes.Kaplan-Meier plotter and GEPIA database performed more analysis and verification on the selected hub genes.Results:In lung adenocarcinoma,the expression of CDK1 was significantly higher than that of normal tissues(P<0.01).UALCAN database analysis showed that clinical characteristics such as stage and gender of lung adenocarcinoma patients were not significantly correlated with the expression of CDK1.GEPIA analysis showed that compared with patients with lung adenocarcinoma with high CDK1 expression,the overall survival rate of patients with low CDK1 expression increased(P<0.05).Oncomine database screened out CDK1 co-expressed genes including TOP2A,MCM4 and other 18 genes.GO analysis showed that CDK1 co-expressed genes were mainly related to chromosome localization,chromosome agglutination,and ATPase activity.KEGG analysis showed that CDK1 co-expressed genes are involved in the p53 signaling pathway and other processes.With String,Cytoscape,and CytoHubba,we obtained the PPI network of CDK1 co-expressed genes,and then selected the hub genes,and selected the first 5 hub genes in the order of Degree,namely CDC6,MAD2L1,TOP2A,KIAA0101,MCM4.Finally,it was confirmed that 5 hub genes are closely related to the prognosis of patients with lung adenocarcinoma.Conclusion:CDK1 is highly expressed in lung adenocarcinoma patients and is closely related to the prognosis of lung adenocarcinoma patients.It provides important guidance for the treatment and diagnosis of lung adenocarcinoma.
