摘要
目的:建立高效液相色谱法同时测定不同豆制品中大豆苷、大豆黄苷染料木苷、染料木素、大豆素和大豆黄素6种大豆异黄酮的含量分析方法。方法:利用高效液相色谱仪,将供试品用80%甲醇溶解提取,用C_(18)色谱柱分离,以乙腈和磷酸水溶液(pH=3)进行梯度洗脱,流速为1.0 mL/min,用紫外检测器在波长260 nm下进行检测。结果:大豆异黄酮各组分在各自范围内线性关系良好。大豆苷、大豆黄苷、染料木苷、大豆素、大豆黄素和染料木素相对标准偏差为0.05%~0.23%,检出限为0.2μg/mL。结论:该方法回收率高,灵敏度高,且操作简单,快速高效,可应用于实际样品中不同种类的大豆异黄酮的分析测定。
Objective:To establish a high performance liquid chromatography method for the simultaneous determination of daidzin,glycitin,genistin,daidzein,daidzein,daidzein(daidzin)in different soybean products.glycitein content analysis method of 6 kinds of soybean isoflavones.Method:Using high performance liquid chromatograph,the test sample was dissolved and extracted with 80%methanol,separated by C_(18) chromatographic column,and eluted with acetonitrile and phosphoric acid aqueous solution(pH=3)for gradient elution at a flow rate of 1.0 mL/min.The detector detects at a wavelength of 260 nm.Result:All components of soybean isoflavones had a good linear relationship within their respective ranges.The relative standard deviations of daidzein,daidzein,genistein,daidzein,daidzein,and genistein were 0.05%~0.23%,and the detection limit was 0.2μg/mL.Conclusion:The method has high recovery rate,high sensitivity,simple operation,rapidity and high efficiency,and can be applied to the analysis and determination of different kinds of soybean isoflavones in actual samples.
