摘要
采用高效凝胶渗透色谱法(HPGPC)以高纯水和硫酸钠溶液作为流动相分别进行样品测定,选择已知分子量的右旋糖酐系列对照品和普鲁兰系列对照品分别进行标准曲线拟合,应用GPC软件分析结果。研究结果表明,不同的色谱条件对海藻酸钠分子量的测定结果有显著差异,以普鲁兰为对照品测得的分子量比用右旋糖酐为对照品测得的结果低;以0.1 mol·L^(-1)硫酸钠溶液为流动相时,海藻酸钠的色谱峰对称性较好。
High performance gel permeation chromatography(HPGPC)was used for the determination of samples with high purity water and sodium sulfate as mobile phase respectively.Dextran standard with known molecular weight and Pullulan polysaccharide standard with known molecular weight were selected for standard curve fitting respectively,and the results were analyzed by GPC softwares.There are significant differences between results of different chromatographic conditions.The molecular weight using pullulan as reference substance is lower than that using dextran as reference substance.When 0.1mol·L^(-1) sodium sulfate solution is used as mobile phase,the symmetry of chromatographic peak of sodium alginate is better.
