摘要
目的研究乳杆菌鼠李糖乳杆菌-格氏乳杆菌联用制剂(LACT)的定植效率,评估其有效剂量在动物体内的急性和长期毒性。方法在食蟹猴中进行2轮阴道定植试验,通过检测阴道分泌物pH值和清洁度,并分离鉴定分泌物中的乳杆菌,确定乳杆菌联用制剂的定植率和定植剂量;通过单次和多次阴道给药,检测联用制剂对食蟹猴体质量、食量、生殖器官及生化指标等的影响,检测联用制剂在体内的急性和长期毒性。结果联用制剂在食蟹猴中连续阴道给药5 d后能成功定植,改善阴道的微生态;甲硝唑预处理能够提高定植效率和定植时间;联用制剂对食蟹猴不具有急性和长期的毒性作用。结论在动物模型中,2种乳杆菌联用制剂对细菌性阴道病具有良好的治疗潜质。
Objective To explore the colonization efficiency of two Lactobacilli(LACT)and evaluate its effective dose,acute and long-term toxicity in animals.Methods Two rounds of vaginal colonization were carried out in Macaca fascicularis(cynomolgus monkey)to detect the pH and cleanliness of vaginal secretions.The bacteria in secretions were isolated and characterized so as to determine the colonization rate and effective dose of LACT.Single and multiple vaginal administration of LACT were performed and the effect of LACT on body weight,food intake,reproductive organs,and some important biochemical parameters were observed,and the acute and long-term toxicity were detected.Results LACT successfully colonized and improved vaginal microecology in cynomolgus monkeys after consecutive vaginal administration for 5 days.Metronidazole pretreatment helped improve the colonization efficiency and duration.Moreover,LACT had no acute or long-term toxic effects on cynomolgus monkeys.Conclusion LACT has good potential for treatment of bacterial vaginosis in the animal model.
