山豆根多糖对PCV2感染小鼠脾淋巴细胞炎症因子mRNA表达的影响 被引量:4

摘要 试验旨在探索山豆根多糖(SSP)对猪圆环病毒2型(PCV2)感染小鼠脾淋巴细胞炎症因子mRNA表达的影响。试验设立细胞对照组、PCV2感染对照组以及PCV2+3种浓度(100、200和400 mg/L)的SSP药物处理组和400 mg/L的SSP药物处理组,共6组。采用PCV2体外感染小鼠脾淋巴细胞,在培养液中加入终浓度为100、200和400 mg/L的SSP,分别培养8、12和24 h后收取样品,荧光定量PCR技术测定细胞内炎性因子的mRNA表达水平。结果显示,当小鼠脾淋巴细胞培养至24 h时,PCV2感染对照组的NF-κB、MCP-1、CXCL10和STAT1的表达水平显著高于细胞对照组(P<0.05);经100、200和400 mg/L的SSP处理后,小鼠脾淋巴细胞炎症因子表达量均有所下降并呈现剂量依赖性,其中PCV2+400 mg/L SSP药物处理组的炎症因子表达量与PCV2感染对照组的相比均差异显著(P<0.05)。研究表明,SSP作用于PCV2感染的小鼠脾淋巴细胞24 h后能够降低细胞内炎症因子的mRNA表达,起到抗炎的效果。 The study was carried out to investigate the effect of Sophora subprosrate polysaccharide(SSP) on mRNA expression levels of inflammatory factors in murine splenic lymphocytes infected with PCV2. The experiment included six groups: Cell control group, PCV2 model group, drug treatment group with different concentrations(PCV2+100, 200,400 mg/L SSP) and 400 mg/L SSP treatment group. Murine splenic lymphocytes were infected with PCV2 in vitro, and then the cells were treated with SSP at 100, 200 and 400 mg/L for 8, 12 and 24 h. The mRNA expression level of inflammatory factors in cells was detected by fluorescence quantitative PCR. The results showed that when the mouse spleen lymphocytes were cultured for 24 h, the expression levels of NF-κB, MCP-1, CXCL10 and STAT1 in the PCV2infection control group were significantly higher than those in cell control group(P<0.05). After treatment with 100, 200and 400 mg/L SSP, the expression of inflammatory factors in mouse spleen lymphocytes decreased in a dose-dependent manner. The expression of inflammatory factors in PCV2 +400 mg/L SSP group was significantly different from that in PCV2 model group(P<0.05). The study indicates that SSP can reduce the mRNA expression of inflammatory factors in PCV2 infected murine splenic lymphocytes for 24 h and play an anti-inflammatory effect.
