摘要
目的 探讨miR-155-5p对MP感染模型大鼠肺功能的保护机制。方法 选取28只清洁及健康的雄性昆明大鼠,随机选取7只分为正常组,剩余21只建立肺炎支原体感染哮喘大鼠,分为模型组、上调组、下调组,各7只。PCR法检测miR-155-5p表达,检测肺功能相关指标、炎症因子相关指标、核因子κB(NF-κB)、信号传导和转录活化因子-3(STAT3)水平。结果 与正常组相比,模型组、上调组、下调组miR-155-5p表达、PAF、IL-13、TNF-α、NF-κB、STAT3水平较高,FVC、FEF25%、FEF50%、FEF75%水平较低(P<0.05);与模型组相比,上调组miR-155-5p表达、PAF、IL-13、TNF-α、NF-κB、STAT3水平较高,FVC、FEF25%、FEF50%、FEF75%水平较低,下调组miR-155-5p表达、PAF、IL-13、TNF-α、NF-κB、STAT3水平较低,FVC、FEF25%、FEF50%、FEF75%水平较高(P<0.05);与上调组相比,下调组miR-155-5p表达、PAF、IL-13、TNF-α、NF-κB、STAT3水平较低,FVC、FEF25%、FEF50%、FEF75%水平较高(P<0.05)。结论 miR-155-5p可能通过调控NF-kB、STAT3通路蛋白水平改善大鼠的肺功能,抑制炎性反应。
Objective rats.MethodsTwenty-eight clean and healthy male Kunming rats were selected,7 were randomly selected and divided into the normal group,and the remaining 21 rats with established Mycoplasma pneumoniae-infected asthma were divided into the model group,the up-regulation group,and the down-regulation group,with 7 rats in each. The expression of miR-155-5p,lung function related indexes,inflammatory cytokine related indexes,Nuclear factor kB(NF-κB),Activator of signal transduction and transcription-3(STAT3)levels were detected by PCR.Results sion of miR-155-5p,PAF,IL-13,TNF-α,NF-κB and STAT3 in the model group,up-regulated group and down-regulated group were higher,FVC,FEF25%,FEF50%,FEF75% The level was lower(P < 0.05);Compared with the model group,the expression of miR-155-5p,PAF,IL-13,TNF-α,NF-κB,and STAT3 in the up-regulated group were higher,while the levels of FVC,FEF25%,FEF50%,and FEF75% were lower in the down-regulated group. The expression of miR-155-5p,PAF,IL-13,TNF-α,NF-κB,and STAT3 levels were lower,and the levels of FVC,FEF25%,FEF50%,and FEF75%were higher(P < 0.05);Compared with the up-regulated group,the down-regulated group had lower miR-155-5p expression,PAF,IL-13,TNF-α,NF-κB,and STAT3 levels,and higher levels of FVC,FEF25%,FEF50%,and FEF75%(P < 0.05).ConclusionThe MiR-155-5p can improve the lung function of rats and inhibit inflammatory response by regulating the protein levels of NF-kB and STAT3 pathways.
