摘要
目的分析跨膜丝氨酸蛋白酶2(TMPRSS2)-成红细胞特异性转化基因相关基因(ERG)、TMPRSS2-成红细胞特异性转化基因变异体1(ETV1)、TMPRSS2-成红细胞特异性转化基因变异体4(ETV4)在前列腺癌中的表达及临床意义。方法2018年4月至2020年9月我院收治的82例前列腺癌患者,采用免疫组化法检测癌组织及癌旁正常组织中TMPRSS2-ERG、TMPRSS2-ETV1、TMPRSS2-ETV4表达。分析上述因子表达与前列腺癌临床病理特征的关系及诊断前列腺癌的价值。结果前列腺癌组织中TMPRSS2-ERG、TMPRSS2-ETV1、TMPRSS2-ETV4阳性表达率均高于癌旁正常组织(P<0.05)。临床分期C+D、Gleason评分≥5分者TMPRSS2-ERG、TMPRSS2-ETV1、TMPRSS2-ETV4阳性表达率高于临床分期A+B、Gleason评分<5分(P<0.05)。RTMPRSS2-ERG、TMPRSS2-ETV1、TMPRSS2-ETV4联合检测曲线下面积(AUC)、敏感度、特异度分别为0.814、0.925、0.865,均高于各项指标单独检测(P<0.05)。结论TMPRSS2-ERG、TMPRSS2-ETV1、TMPRSS2-ETV4在前列腺癌患者中呈过度表达,通过联合检测上述因子对前列腺癌的诊断与鉴别诊断有良好的应用价值。
Objective To analyze the expression and clinical significance of transmembrane serine protease 2(TMPRSS2)-erythroblast specific transformation genes and related genes(ERG),TMPRSS2-erythroblast specific transformation gene variant 1(ETV1)and TMPRSS2-erythroblast specific transformation gene variant 4(ETV4)in prostate cancer.Methods Clinical data of 82 prostate cancer patients admitted to our hospital from April 2018 to September 2020 were collected.Immunohistochemistry was used to detect the expression of TMPRSS2-ERG,TMPRSS2-ETV1,and TMPRSS2-ETV4 in cancer tissues and normal adjacent tissues.The relationship between expressions of the above factors and clinicopathological characteristics of prostate cancer was analyzed.The diagnostic value of the factors for prostate cancer were evaluated.Results The positive expression rates of TMPRSS2-ERG,TMPRSS2-ETV1,and TMPRSS2-ETV4 in prostate cancer tissues were significantly higher than those in normal tissues adjacent to cancer(P<0.05).The positive expression rates of TMPRSS2-ERG,TMPRSS2-ETV1,and TMPRSS2-ETV4 in clinical stage C+D and Gleason score≥5 were significantly higher than those in clinical stage A+B and Gleason score<5(P<0.05).The ROC curve analysis showed that the area under curve(AUC),sensitivity,and specificity of the combined detection of the three indexes were 0.814,0.925,and 0.865,respectively,which were all higher than those of individual detection.Conclusion TMPRSS2-ERG,TMPRSS2-ETV1 and TMPRSS2-ETV4 are over expressed in prostate cancer patients.The combined detection of these factors has a good application value in the diagnosis and differential diagnosis of prostate cancer.
