摘要
介绍了一种能同时测定DNA中鸟嘌呤(G),腺嘌呤(A),胸腺嘧啶(T)和胞嘧啶(C)的新型的电化学修饰电极。采用循环伏安法将钙指示剂修饰到玻碳电极表面,通过示差脉冲伏安法(DPV)进行检测。和裸电极相比,钙指示剂修饰电极有良好的电化学催化活性,对嘌呤和嘧啶类的氧化信号十分显著。此外,该方法已成功应用于测定鲑鱼精DNA中G,A,T和C的含量,结果令人满意。
A new type of electrochemical modified electrode was developed for the simultaneous determination of guanine(G),adenine(A),thymine(T)and cytosine(C)in DNA.The calcon-carboxylic acid modified glassy carbon electrode was prepared by using cyclic voltammetric method.Compared to the bare electrode,the modified electrode exhibited remarkable electrocatalytic activities towarded the oxidation of G,A,T and C due to the advantages of high electrode activity.In addition,the proposed method was successfully applied to the assessment of G,A,T and C contents in a salmon sperm DNA sample with satisfactory results.
