摘要
目的使用同位素标记相对和绝对定量(isobaric tags for relative and absolute quantitation,iTRAQ)蛋白组学技术分析男性痛风患者和正常对照组尿液中的差异蛋白质特征谱,为今后进一步研究打下基础。方法收集男性痛风患者和体检中心健康男性尿液标本各6例,应用iTRAQ技术对样本进行鉴定,对获得的全部蛋白进行生物信息学分析。结果(1)两组共筛选出1980个蛋白。取差异倍数≥1.5或≤0.67为筛选标准,共筛选出差异蛋白226个,其中上调差异蛋白120个,下调差异蛋白106个;(2)基于本体论(gene ontology,GO)分析显示差异蛋白主要参与炎症反应、免疫反应和对细菌的防御反应;(3)KEGG(Kyoto Encyclopedia of Genes and Genomes)通路分析差异蛋白主要富集在甘油磷脂代谢、肾素-血管紧张素系统和矿物吸收三个代谢途径。结论痛风组与正常组对差异表达蛋白进行KEGG信号通路富集分析,发现主要富集在脂代谢、肾素-血管紧张素系统和矿物吸收三条分子信号通路。
Objective To analyze differential protein map between male gout patients and normal controls with isotope-labeled relative and absolute quantification(iTRAQ)technology in proteomics,so as to laid the foundation for the future research.Methods Urinary samples were collected from 6 cases of male patients with gout and 6 cases of healthy males.After extraction process,the differential proteins were identified by iTRAQ.The analysis was conducted using bioinformatics software.Results(1)A total of 1980 proteins were identified in the two groups.In reference to the screening criteria,the differential multiples≥1.5 or≤0.67,a total of 226 differential proteins were identified.Among them,the expressions of 120 proteins were up-regulated and 106 proteins down-regulated.(2)Based on the gene ontology(GO),it was indicated that the differential proteins were enriched in the inflammatory response,immune response,defense response to bacterium,etc.(3)The analysis of KEGG(Kyoto encyclopedia of genes and genomes)pathway showed that the differential proteins were mainly related to glycerophospholipid metabolism,renin-angiotensin system and mineral absorption.Conclusion KEGG signal pathway enrichment analysis of differentially expressed proteins between male gout patients and normal controls show that they were mainly enriched in three molecular signal pathways:glycerophospholipid metabolism,renin-angiotensin system and mineral absorption.
