摘要
高分辨率熔解曲线技术(HRM)作为近年来新兴的PCR分析技术,快捷简便、分辨率高。已克隆的稻瘟病抗性主效基因Pi2,在水稻中表现出稳定的稻瘟病抗性,目前已广泛用于稻瘟病抗性育种中。为明确该基因在黑龙江省种质资源中的分布情况,本研究利用Pi2的HRM功能型分子标记,在67份部分黑龙江省主栽品种及优异种质资源中进行基因分型。结果表明,67份种质中,含纯合pi2有9份,占参试种质的13.43%,分别为龙粳12、哈99774、龙粳31、龙粳39、龙粳54、龙交10-989、龙粳53、彩及zlm55-1。
HRM(high resolution melting)technology as a new PCR analysis technology in recent years,with the characters of simple,fast and high resolution,is widely concerned by the broad masses of the researchers.As the cloned main genes of rice blast resistance,Pi2 have been widely used in rice blast resistance breeding for their stable resistance.In order to determine the distribution of the two gene in germplasm resources of Heilongjiang Province,67 main cultivated varieties and germplasm resources of Heilongjiang Province were genotyped by the HRM(high resolution melting)-based functional markers of pi2.The results showed that,9 germplasm resources contained the homozygous gene pi2(13.43%),which are Longgeng-12,Ha-99774,Longgeng-31,Longgeng-39,Longgeng-54,Longjiao10-989,Longgeng-53,Cai and Zlm55-1.
