药物血脑屏障通透性体外模型及评价方法的研究进展 被引量:1

摘要 血脑屏障(BBB)是位于循环血液与中枢细胞外液之间的一道天然生理屏障,其表现出的动态选择性滤过作用决定了BBB在防卫中枢神经系统(CNS)免受异物侵害的同时也限制了相关药物的递送吸收。本文通过查阅近年来国内外相关文献,梳理常见BBB体外细胞模型的优缺点,并对各种药物BBB通透性方法进行评价分析,揭示构建一种高度契合人体内真实BBB条件的模型,不仅对BBB生理机制进行研究,而且在新型靶向药物的设计与筛选中都具有极大帮助。最后展望当前中药纳米递药系统这一热点研究方向,为深入开发穿透BBB治疗CNS疾病的药物提供积极探索。 The blood-brain barrier(BBB)is a natural physiological barrier located between circulating blood and central extracellular fluid.The demonstrated dynamic selective filtering function determines that it protects the central nervous system(CNS)from foreign bodies while also limiting the delivery and absorption of relevant drugs.In this paper,the advantages and disadvantages of common BBB cell models in vitro are summarized by reviewing relevant literatures at home and abroad in recent years,and the BBB permeability methods of various drugs are evaluated and analyzed,revealing that building a model that highly fits the real BBB conditions in humans is not only greatly helpful in the study of BBB physiological mechanisms,moreover,in the design and screening of novel targeted drugs.Finally,expecting to this hot research field of traditional Chinese medicine nanocarrier-based delivery system,which provides active exploration for the in-depth development of drugs passing through the BBB for the treatment of CNS diseases.
