摘要
目的人脐带间充质干细胞(hUMSCs)外泌体在保护人视网膜色素上皮(RPE)细胞抵抗高糖诱导的细胞凋亡中的作用。方法实验研究。超速离心法收集hUMSCs外泌体,通过透射电镜和纳米颗粒追踪分析其形态结构;采用CCK-8和EdU实验检测外泌体处理后的人RPE在高糖条件下的增殖情况;通过蛋白免疫印迹(western blot)检测周期蛋白和凋亡蛋白的变化;运用流式细胞仪检测hUMSCs外泌体在高糖诱导凋亡率中的作用。结果高浓度糖培养环境通过促进线粒体介导的细胞凋亡而抑制了ARPE-19的增殖;来自hUMSCs的外泌体(hUMSC-exo)能促进人RPE细胞的增殖及保护ARPE-19抵抗高糖诱导的细胞凋亡。结论hUMSCs外泌体能够保护人RPE细胞抵抗高糖诱导的细胞凋亡。
Objective To explore the role of exosomes that from human umbilical cord mesenchymal stem cells in protecting human pigment retinal epithelial cells against apoptosis induced by high glucose.Methods Exosomes of human umbilical cord mesenchymal stem cells were collected by ultracentrifugation,and their morphology and structure were analyzed by transmission electron microscopy and Nanoparticle Tracking Analysis.CCk-8 and EdU experiments were used to detect the proliferation of human pigment retinal epithelial cells after treating with exosomes under high glucose conditions.The changes of cyclin and apoptotic proteins were detected by Western blot.Flow cytometry was used to detect the role of exosomes from mesenchymal stem cells in apoptosis rate induced by high glucose.Results High concentration of sugar inhibited the proliferation of ARPE-19 by promoting mitochondria-mediated apoptosis.Exosomes(HUMSC-Exo)from hUMSCs can promote the proliferation of human retinal pigment epithelial cells and protect ARPE-19 against apoptosis induced by high glucose.Conclusions Exosomes from human umbilical cord mesenchymal stem cells can protect human pigment retinal epithelial cells against apoptosis that induced by high glucose.
