鸢尾甲苷B对氧糖剥夺/复氧损伤PC12细胞的神经保护作用 被引量:1

Neuroprotective Effect of Iristectorin B in the Shoot Stem on PC12 Cells Injured by Oxygen Glucose Deprivation/Reoxygenation

摘要 利用PC12细胞建立氧糖剥夺(OGD)体外脑卒中损伤模型,采用该模型对射干中鸢尾甲苷B抗脑卒中进行活性评价。评价指标包括:细胞形态学观察、细胞存活率检测、细胞内活性氧(ROS)含量检测、乳酸脱氢酶(LDH)释放率测定、细胞内钙离子(Ca^(2+))浓度检测、细胞凋亡情况检测等。结果显示,射干中天然异黄酮鸢尾甲苷B能显著减轻氧糖剥夺/复氧(OGD/R)造成的PC12细胞损伤,降低细胞凋亡,提升细胞存活率,降低Ca^(2+)、LDH、ROS水平。结果表明,鸢尾甲苷B对Na_(2)S_(2)O_(4)损伤的PC12细胞有明显的保护作用,其机理可能与脑内神经元的保护作用相关。 PC12 cells were used to establish an in vitro stroke injury model of glucose oxygen deprivation(OGD),and the activity of Iristectorin B in the ejaculatory trunk was evaluated against stroke.The evaluation indexes included the following:cell morphology observation,cell survival rate detection,intracellular reactive oxygen species(ROS)content detection,lactate dehydrogenase(LDH)release rate measurement,intracellular calcium ion(Ca^(2+)) concentration detection,apoptosis situation detection and so on.The results showed that Iristectorin B,a natural isoflavone present in the shoot trunk,significantly attenuated PC12 cell injury caused by oxygen glucose deprivation/reoxygenation(OGD/R),decreased cell apoptosis,elevated cell survival,and decreased Ca^(2+),LDH,and ROS levels.The results showed that Iristectorin B had obvious protective effect on Na_(2)S_(2)O_(4) injured PC12 cells,and the mechanism may be associated with the protective effect of neurons in the brain.
