摘要 目的:探究长链非编码核糖核酸LOC100505851(lncRNA-LOC100505851)对肝细胞癌(HCC)的诊断及预后判断价值。方法:选取接受肝脏切除手术的HCC患者80例作为研究对象。实时荧光定量聚合酶链式反应(qRT-PCR)检测癌组织及癌旁组织的lncRNA-LOC100505851水平,绘制受试者工作特征(ROC)曲线评价组织lncRNA-LOC100505851表达水平对HCC的诊断效能。根据癌组织lncRNA-LOC100505851表达水平分为低表达组与高表达组,分析两组患者的临床病理特征与预后差异。结果:lncRNA-LOC100505851在HCC组织中的表达水平低于对应的癌旁组织(P<0.05);癌组织lncRNA-LOC100505851水平诊断HCC的ROC曲线下面积(AUC)为0.863(95%CI=0.808~0.918,P<0.05),最佳截断值为0.695;按照最佳截断值将lncRNA-LOC100505851表达分为高表达组和低表达组,分析其与临床病理特征的关系,结果显示,HCC组织中lncRNA-LOC100505851表达与肿瘤大小、淋巴结转移及TNM分期密切相关(P<0.05);生存分析结果显示,lncRNA-LOC100505851低表达组第1、2、3年的总生存率及无瘤生存率均低于lncRNA-LOC100505851高表达组;单因素分析结果显示,肿瘤大小≥5 cm,TNM分期为Ⅲ~Ⅳ期,淋巴结转移与lncRNA-LOC100505851低表达为HCC患者总生存时间的独立危险因素(P<0.05);多因素分析结果显示,肿瘤大小≥5 cm,TNM分期为Ⅲ~Ⅳ期与lncRNA-LOC100505851低表达为HCC患者总生存时间的独立预测因子(P<0.05)。结论:lncRNA-LOC100505851的表达与HCC患者的临床病理特征密切相关,可作为HCC患者预后评估和治疗的潜在靶点。 Objective:To explore the diagnostic and prognostic value of long non-coding RNA LOC100505851(lncRNA-LOC100505851)for hepatocellular carcinoma(HCC).Methods:80 HCC patients who underwent liver resection were selected as the study subjects.The levels of lncrNA-Loc100505851 in cancer tissues and adjacent tissues of the selected subjects were detected by qRT-PCR,and ROC curves were drawn to evaluate the diagnostic efficacy of tissue lncrNA-Loc100505851 expression level for HCC.According to the expression level of lncrNA-LOC100505851 in cancer tissues,the patients were divided into the low expression group and the high expression group,and the clinicopathological characteristics and prognosis of the two groups were analyzed.Results:The expression level of lncrNA-LOC100505851 in HCC tissues was significantly lower than that in adjacent tissues(P<0.005).The area under ROC curve for HCC diagnosis at the level of cancer tissue lncrNA-LOC100505851 was 0.863(95%CI=0.808-0.918,P<0.05),the optimal truncation value was 0.695.According to the optimal truncation value,lncrNA-Loc100505851 expression was divided into high expression group and low expression group,and the relationship between lncrNA-Loc100505851 expression and clinicopathological characteristics was analyzed.The results showed that the expression of LNcrNA-Loc100505851 in HCC tissues was significantly correlated with tumor size,lymph node metastasis and TNM stage(P<0.05).Survival analysis showed that the overall survival rate and disease-free survival rate at 1,2,and 3 years in the low-expression lncrNA-LOC100505851 group were lower than those in the high-expression LNcrNA-LOC100505851 group.Univariate analysis showed that tumor size≥5 cm,TNM stageⅢ~Ⅳ,lymph node metastasis and low expression of lncrNA-LOC100505851 were risk factors for overall survival of HCC patients(P<0.05).Multivariate analysis showed that tumor size≥5 cm,TNM stageⅢ~Ⅳand low expression of LNcrNA-LOC100505851 were independent predictors of overall survival of HCC patients(P<0.05).Conclusion:The expression of lncRNA-LOC100505851 is closely related to the clinicopathological characteristics of HCC patients,which maybe be used as a potential target for prognosis evaluation and treatment of HCC patients.
