益气化瘀解毒方拆方对急性呼吸窘迫综合征大鼠的预防作用研究

Preventive Effect of Various Treatments of Yiqi Huayu Jiedu Decoction on Acute Respiratory Distress Syndrome Rats

目的 探讨益气化瘀解毒方拆方对急性呼吸窘迫综合征大鼠的抗炎作用及其对TLR4-NLRP3通路的影响。方法 将30只雄性SD大鼠随机等分为5组,空白对照组、模型组、益气解毒组、益气化瘀组、益气逐饮组。其中各中药治疗组给予对应的中药灌胃治疗,模型组和各中药治疗组以尾静脉注射脂多糖方法,制备全身炎症反应所致的急性呼吸窘迫综合征大鼠模型。观察各组大鼠肺组织病理变化、采用酶联免疫法检测肺组织、肺泡灌洗液中TLR4-NLRP3通路相关蛋白表达水平和血清中炎性因子白细胞介素-1β(IL-1β)、白细胞介素-18(IL-18)蛋白表达水平。结果 模型组肺组织表现符合急性呼吸窘迫综合征大鼠肺组织病理特征,各中药治疗组肺组织炎性损伤程度较模型组降低。与模型组相比,各治疗组可显著减少大鼠肺匀浆中TLR4表达水平和肺泡灌洗液中NLRP3表达水平。各治疗组均可显著减少大鼠血清炎性因子IL-1β、IL-18表达水平,其中益气解毒组效果最佳。结论 益气化瘀解毒方各拆方组均可以减轻大鼠肺组织损伤,减少TLR4-NLRP3通路相关蛋白及血清中炎性因子表达水平,TLR4-NLRP3通路可能是其发挥抗炎作用干预ARDS的重要机制之一。

Objective:To investigate the preventive effect of various treatments of Yiqi Huayu Jiedu Decoction on acute respiratory distress syndrome(ARDS)and its influence on TLR4-NLRP3 pathway in rats. Methods:A total of 30 male SD rats were randomly divided into 5 groups:blank control group,model group,Yiqi Jiedu group,Yiqi Huayu group and Yiqi Zhuyin group. Each Chinese medicine treatment group was given the corresponding Chinese medicine intragastric treatment,and the model group and each Chinese medicine treatment group were injected with lipopolysaccharide through tail vein to prepare the rat model of ARDS caused by systemic inflammation. The pathological changes of lung tissues were observed,and the expression levels of TLR4-NLRP3 pathwayrelated proteins in lung tissues and alveolar lavage fluid and the expression levels of inflammatory factors IL-1βand IL-18 in serum were detected by ELISA. Results:The lung tissue of the model group was consistent with the pathological characteristics of the lung tissue of the rats with ARDS. The inflammatory injury degree of the lung tissue of the Chinese medicine treatment groups was lower than that of the model group. Compared with model group,TLR4 expression level in lung homogenate and NLRP3 expression level in alveolar lavage fluid of rats were significantly reduced in all treatment groups. All treatment groups can significantly reduce the expression levels of inflammatory factors IL-1β and IL-18,among which Yiqi Jidu group has the best reduction effect. Conclusion:Yiqi Huayu Jiudu Decoction can alleviate lung tissue injury in rats and reduce the expression levels of TLR4-NLRP3 pathway related proteins and inflammatory factors in serum. TLR4-NLRP3 pathway may be one of the important mechanisms of its anti-inflammatory effect on ARDS.
