参芪益心方对心力衰竭大鼠心功能及其心肌细胞凋亡率的影响 被引量:3

Effects of Shenqi Yixin Decoction on Cardiac Function and Cardiomyocyte Apoptosis Rate in Rats with Heart Failure

摘要 目的 观察参芪益心方对心衰大鼠心功能与心肌细胞凋亡率的影响,研究参芪益心方对心衰大鼠心室重塑的机制与效果。方法 腹腔注射阿霉素建立心衰大鼠模型,将90只大鼠造模后随机分为正常对照组(自由饮食)、模型组(生理盐水10 m L/kg)、西药对照组(卡维地洛混悬液0.558 mg/kg)、芪苈强心组(芪苈强心胶囊混悬液0.3214 g/kg)、参芪益心方低剂量组(1.696 g/mL)、参芪益心方高剂量组(3.392 g/mL)。超声心动图检测心功能;CMIAS法对细胞凋亡切片进行分析来观察心肌细胞凋亡密度。结果 与模型组相比,各治疗组的心功能指数均有改善(P <0.01);参芪益心方低剂量组与西药对照组、芪苈强心组相比较,各指标都有显著改善(P <0.01)。与其他组相比,参芪益心方高剂量组的凋亡细胞表面密度明显减少(P <0.01)。结论 参芪益心方可以增强心衰模型大鼠的心功能,减轻建模大鼠的心肌损伤。 Objective:To observe the effects of Shenqi Yixin Decoction on cardiac function and apoptosis rate of cardiomyocytes in heart failure rats,and to investigate the mechanism and its effect on ventricular remodeling in rats with heart failure. Methods:Rat model of heart failure induced by intraperitoneal injection of doxorubicin was established. After establishment of 90 rats in sequence,the rats were randomly divided into normal control groups(free diet and water),model group(saline infusion 10 m L/kg),western medicine(carvedilol suspension0.558 mg/kg) control group,traditional Chinese medicine(Astragalus hebecarbarum heart capsule suspension0.3214 g/kg)control group,low dose group of Chinese medicine(1.696 g/mL),and high dose group of Chinese medicine(3.392 g/mL)after modeling. Cardiac function was measured by echocardiography and the multi-functional true color pathology image analysis system CMIAS was used to analyze the TUNEL method of apoptosis section to observe the density of apoptosis in cardiac myocytes. Results:Cardiac function improved in all treatment groups compared to the model group(P < 0.01);the low-dose group of Chinese medicine showed significant improvement in the values of all indices compared with the western control group and the Chinese medicine control group(P < 0.01). The surface density of apoptotic cells was significantly reduced in the herbal high-dose group compared with the rest of the groups(P < 0.01). Conclusion:Shenqi Yixin Decoction can enhance cardiac function and reduce myocardial injury in modeled rats.
