摘要
为了改进山羊育种,提供有用的遗传标记,首先对3个山羊品种(云上黑山羊,济宁青山羊,辽宁绒山羊)768只母羊PARD3B和ESYT1基因的8个候选位点进行基因分型,之后再进行群体遗传学分析和关联分析。结果表明,3个山羊品种有7个候选位点均存在多态性,1个候选位点ESYT1 g.56562540C>G不存在多态性。PARD3B基因g.42671330T>C位点和g.42147569C>G位点在3个群体中均为中度多态性(0.25<PIC<0.5);g.42423542C>T位点在辽宁绒山羊和云上黑山羊群体中为低度多态性(PIC<0.25),而在济宁青山羊群体中则为高度多态性(PIC>0.5);g.41961765C>T位点在济宁青山羊和云上黑山羊群体中均为低度多态性(PIC<0.25),在辽宁绒山羊群体中为中度多态性(0.25<PIC<0.5);g.41961457C>T位点和g.41961508A>G位点在辽宁绒山羊和济宁青山羊中均不存在多态性,在云上黑山羊中分别为中度多态性(0.25<PIC<0.5)和低度多态性(PIC<0.25)。ESYT1基因g.56567828T>C位点在3个群体中均为低度多态性(PIC<0.25)。卡方检验表明,大部分位点均处于Hardy-Weinberg平衡状态(P>0.05)。关联分析表明,在云上黑山羊中,PARD3B基因的g.41961457C>T位点TT型个体的产羔数显著高于CC型个体(P<0.05);g.41961508A>G位点GA型个体的断奶窝重显著高于AA型个体(P<0.05);g.42423542C>T位点TT型个体的断奶窝重显著高于CT型个体(P<0.05)。综上所述,PARD3B基因的g.41961457C>T位点可以作为云上黑山羊产羔数选择的潜在分子标记,g.41961508A>G位点和g.42423542C>T位点可作为云上黑山羊断奶窝重选择的潜在分子标记。
In order to improve goat breeding and provide useful genetic markers,this study first genotyped 8 candidate loci of PARD3B and ESYT1 genes in 768 ewes of three goat breed(Yunshang Black goats,Jining Grey goats,Liaoning Cashmere goats),and then carried out population genetic and association analysis.The results showed that seven candidate loci were polymorphic,and ESYT1 g.56562540C>G was not polymorphic.The loci of g.42671330T>C and g.42147569C>G of PARD3B gene were moderately polymorphic in all three populations(0.25<PIC<0.5);g.42423542C>T was low polymorphism(PIC<0.25)in Liaoning Cashmere goats and Yunshang Black goats population,but highly polymorphic(PIC>0.5)in Jining Grey goats population;g.41961765C>T was moderately polymorphic in Liaoning Cashmere goat population(0.25<PIC<0.5);g.41961457C>T and g.41961508A>G had no polymorphism in Liaoning Cashmere goats and Jining Grey goats,but moderate polymor-phism and low polymorphism in Yunshang Black goats;the g.56567828T>C locus of ESYT1 gene was low polymorphic in all three populations(PIC<0.25).Chi-square test showed that most of the loci were in Hardy-Weinberg equilibrium(P>0.05).Association analysis showed that in Yunshang Black goats,the litter weight of individuals with GT genotype at g.41961457C>T locus of PARD3B gene was significantly higher than that of CC genotype(P<0.05),the weaning litter weight of individuals with GA genotype at g.41961508A>G locus was significantly higher than that of AA type(P<0.05),and the weaning litter weight of individuals with GT genotype at g.42423542C>T locus was significantly higher than that of CT type(P<0.05).To sum up,a locus g.41961457C>T in PARD3B gene can be used as a potential molecular marker for litter size selection of Yunshang Black goats,and g.41961508A>G and g.42423542C>T can be used as potential molecular markers for litter weight selection of Yunshang Black goats.
