红芪多糖通过Wnt通路减轻脂多糖诱导牙髓细胞凋亡的作用及机制 被引量:3

Effect and mechanism of Hedysari polysaccharide on reducing tlipopolysaccharide induced apoptosis of dental pulp cells through Wnt pathway.

摘要 目的:研究红芪多糖(HSP)通过Wnt通路减轻脂多糖(LPS)诱导牙髓细胞凋亡的作用及机制。方法:培养人牙髓细胞,分组并用10 mg/L LPS、10 mg/L或100 mg/L HSP、0.5 mg/L DKK1处理,采用MTS法检测细胞存活率,TUNEL法检测细胞凋亡率,Western Blot检测Bax、Cleaved caspase-3、Wnt1、β-catenin的表达水平。结果:LPS组牙髓细胞的存活率及Wnt1、β-catenin的表达水平低于对照组,凋亡率及Bax、Cleaved caspase-3的表达水平高于对照组;LPS+10 mg/L HSP组及LPS+100 mg/L HSP组牙髓细胞的存活率及Wnt1、β-catenin的表达水平高于LPS组,凋亡率及Bax、Cleaved caspase-3的表达水平低于对LPS组;DKK1+LPS+100 mg/L HSP组牙髓细胞的存活率低于溶剂+LPS+100 mg/L HSP组,凋亡率及Bax、Cleaved caspase-3的表达水平高于溶剂+LPS+100 mg/L HSP组。结论:HSP通过激活Wnt通路减轻LPS诱导牙髓细胞凋亡。 Objective:To study the effect and mechanism of Hedysari polysaccharide(HSP)on reducing the apoptosis of dental pulp cells induced by lipopolysaccharide(LPS)through Wnt pathway.Methods:Human dental pulp cells were cultured and treated with 10 mg/L LPS,10 mg/L or 100 mg/L HSP,0.5 mg/L DKK1.The cell survival rate was detected by MTS method,the apoptosis rate was detected by TUNEL method,and the expression levels of Bax,cleaved Caspase-3,Wnt1 andβ-catenin were detected by Western Blot.Results:The survival rate of dental pulp cells and the expression levels of Wnt1 andβ-catenin in LPS group were lower than those in control group,while the apoptosis rate and the expression levels of Bax and cleaved caspase-3 were higher than those in control group;the survival rate and the expression levels of Wnt1 andβ-catenin in LPS+10 mg/L HSP group and LPS+100 mg/L HSP group were higher than those in LPS group,and the apoptosis rate and the expression levels of Bax and cleaved caspase-3 were lower than those in LPS group.The survival rate of DKK1+LPS+100 mg/L HSP group was lower than that in solvent+LPS+100 mg/L HSP group,the apoptosis rate and the expression levels of Bax and cleaved caspase-3 were higher than those in olvent+LPS+100 mg/L HSP group.Conclusion:HSP attenuates LPS induced apoptosis of dental pulp cells by activating Wnt pathway.
