摘要
目的 观察五味子联合阿卡波糖对2型糖尿病小鼠胰岛素抵抗及降血糖作用的机制。方法 采用链脲佐菌素(STZ)腹膜内注射建立2型糖尿病小鼠模型,8周龄雄性C57BL/6J小鼠50只分为正常组、模型组、五味子组、阿卡波糖组、联合组5组,每组10只,治疗4周后,测定空腹胰岛素(FINS)、胰岛素抵抗指数(HOMA-IR)、β细胞功能(HOMA-β)、胰岛素敏感性检测指数(QUICKI)、空腹血糖(FPG)、白细胞介素-4(IL-4)、白细胞介素-8(IL-8)、肿瘤坏死因子-α(TNF-α)及磷酸肌醇-3-激酶(PI3K)、蛋白激酶B(Akt)mRNA。结果 与正常组比较,模型组FINS、FPG、HOMA-IR、IL-4、IL-8、TNF-α水平升高,HOMA-β、QUICKI、胰腺PI3K、AktmRNA和蛋白相对表达量降低(P<0.05);与模型组比较,五味子组、阿卡波糖组、联合组空腹FINS、FPG、HOMA-IR、IL-4、IL-8、TNF-α水平降低,HOMA-β、QUICKI、胰腺PI3K、AktmRNA蛋白相对表达量升高(P<0.05);与五味子组、阿卡波糖组比较,联合组FINS、FPG、HOMA-IR、IL-4、IL-8、TNF-α水平降低,HOMA-β、QUICKI、胰腺PI3K、AktmRNA和蛋白相对表达量升高(P<0.05)。结论 五味子联合阿卡波糖能改善对2型糖尿病小鼠胰岛素抵抗作用,降低2型糖尿病小鼠血糖,降低炎症因子水平;其机制可能与五味子联合阿卡波糖能促进2型糖尿病小鼠鼠胰腺PI3K、Akt表达有关。
Objective To observe the mechanism of Schisandra chinensis combined with acarbose on insulin resistance and hypoglycemic effect in type 2 diabetic mice.Methods A mouse model of type 2 diabetes was established by intraperitoneal injection of streptozotocin(STZ).Fifty 8-week-old male C57 BL/6 J mice were divided into normal group,model group Schisandra group acarbose group and combined group.There were 5 groups of 10 animals in each group.After 4 weeks of treatment,insulin resistance and blood glucose(fasting insulin(FINS),insulin resistance index(HOMA-IR),β-cell function(HOMA-β),insulin sensitivity detection index(QUICKI),fasting plasma glucose(FPG) levels pancreatic tissue inflammatory factors interleukin(IL)-4,IL-8,tumor necrosis factor-α(TNF-α) proteins levels and signaling pathways [phosphoinositide-3-kinase(PI3 K),protein kinase B(Akt) mRNA and protein] levels were measured.Results Compared with normal group,the levels of FINS,FPG,HOMA-IR,IL-4,IL-8 and TNF-α in model group were increased,while the levels of HOMA-β,QUICKI pancreatic PI3 K,Akt mRNA and protein relative expression were decreased(P<0.05);compared with model group,the levels of FINS,FPG,HOMA-IR, IL-4, IL-8 and TNF-α in Schisandra group,acarbose group and combination group were decreased,and the levels of HOMA-β,QUICKI,pancreatic PI3 K,Akt mRNA and protein relative expression were decreased;compared with Schisandra group and acarbose group,the levels of FINS, FPG,HOMA-IR, IL-4,IL-8 and TNF-α in combination group were decreased,and the levels of HOMA-β,QUICKI,pancreatic PI3 K,Akt mRNA and protein relative expression were increased(P<0.05).Conclusion Schisandra chinensis combined with acarbose could improve insulin resistance in type 2 diabetic mice and reduce blood sugar in type 2 diabetic mice;the mechanism might be that Schisandra chinensis combined with acarbose could promote the expression of PI3 K and Akt in the pancreas of type 2 diabetic mice.
