急性心肌梗死患者血清中miR-206、Cx43表达及与预后的关系 被引量:4

Expression of miR-206 and Cx43 in Serum of Patients with Acute Myocardial Infarction and Their Relationship with Prognosis

摘要 目的探究急性心肌梗死(AMI)患者血清中微小RNA-206(miR-206)和细胞间隙连接蛋白43(Cx43)表达水平及与预后的关系。方法选取2018年11月至2020年11月收治的190例AMI患者,根据是否发生心血管事件,将其分为心血管事件组(57例)和无心血管事件组(133例)。采用实时荧光定量PCR(qRT-PCR)法检测血清miR-206表达水平;采用酶联免疫吸附法(ELISA)检测血清Cx43表达水平。利用受试者操作特征曲线分析血清miR-206、Cx43表达水平对AMI后发生心血管事件的预测价值;利用多因素Logistic回归分析影响AMI后发生心血管事件的危险因素。结果心血管事件组患者左室射血分数(LVEF)及血清Cx43表达水平明显低于无心血管事件组(P<0.05),血清肌钙蛋白I(cTnI)、肌酸激酶同工酶(CKMB)、miR-206表达水平明显高于无心血管事件组(P<0.05)。血清miR-206联合Cx43表达水平预测AMI后发生心血管事件的曲线下面积为0.920,明显高于血清miR-206、Cx43表达水平单独预测(P<0.05)。miR-206、Cx43是AMI后发生心血管事件的独立危险因素(P<0.05)。结论血清miR-206表达水平升高、血清Cx43表达水平降低与AMI后心血管事件的发生有关,均可作为评估AMI后心血管事件的血清指标。 Objective To explore the relationship between the expression levels of microRNA-206(miR-206)and connexin 43(Cx43)in the serum of patients with acute myocardial infarction(AMI)and their prognosis.Methods A total of 190 patients with AMI treated in our hospital from November 2018 to November 2020 were selected and divided into cardiovascular event group(57 cases)and non cardiovascular event group(133 cases)according to whether cardiovascular events occurred.The expression level of serum miR-206 was detected by quantitative real-time PCR(qRT-PCR),the expression of Cx43 in serum was detected by enzyme linked immunosorbent assay(ELISA).Receiver operating characteristic(ROC)curve was used to analyze the predictive value of serum miR-206 and Cx43 expression levels for cardiovascular events after AMI,multivariate Logistic regression was used to analyze the risk factors of cardiovascular events after AMI.Results The expression levels of left ventricular ejection fraction(LVEF)and serum Cx43 in the cardiovascular event group were significantly lower than those in the non cardiovascular event group(P<0.05),and the expression levels of serum cardiac troponin I(cTnI),creatine kinase isoenzyme(CK-MB)and miR-206were significantly higher than those in the non cardiovascular event group(P<0.05).The area under the curve of serum miR-206 combined with Cx43 expression level in predicting cardiovascular events after AMI was 0.920,which was significantly higher than that predicted by serum miR-206and Cx43 expression level alone(P<0.05).miR-206 and Cx43 were independent risk factors for cardiovascular events after AMI(P<0.05).Conclusion The increase of serum miR-206 expression level and the decrease of serum Cx43 expression level are related to the occurrence of cardiovascular events after AMI,which can be used as serum indexes to evaluate cardiovascular events after AMI.
