电针联合跑轮训练对MCAO模型大鼠自噬、神经细胞凋亡和神经发生的影响

Effect of Electroacupuncture Combined with Running Wheel Training on Nerve Cell Autophagy,Apoptosis and Neurogenesis in MCAO Rat Model

摘要 目的 探究电针联合跑轮训练对大脑中动脉栓塞(middle cerebral artery embolism,MCAO)模型大鼠自噬、神经细胞凋亡和发生的影响.方法 30只SD大鼠随机分为MCAO模型组、联合干预组和假手术组,每组10只.测量神经系统评分和脑梗塞体积.检测自噬效应蛋白(Beclin1)和膜型微管相关蛋白1A/1B-轻链3(LC3)(LC3B-Ⅱ)/胞浆型微管相关蛋白1A/1B-轻链3(LC3)(LC3B-Ⅰ)表达以及凋亡相关蛋白B淋巴细胞瘤-2基因(bcl-2)、BCL2关联X蛋白(BAX)和半胱氨酸蛋白酶-3(Caspase-3)表达.检测神经细胞凋亡和超氧化物歧化酶(SOD)和过氧化氢酶(CAT)的活性以及丙二醛(MDA)、p-AMPK和p-mTOR的表达.结果 MCAO模型组Tunel阳性细胞、神经系统评分、脑梗塞体积、BAX、Caspase-3、Beclin-1、LC3B-Ⅱ/LC3B-I、p-AMPK表达、MDA含量增加,SOD和CAT活性、p-mTOR、Bcl-2表达降低(P<0.05).联合干预组Tunel阳性细胞、神经系统评分、脑梗塞体积、BAX、Caspase-3、Beclin-1和LC3B-Ⅱ/LC3B-I、p-AMPK表达、MDA含量降低,SOD和CAT活性、Bcl-2、p-mTOR表达增加(P<0.05).结论 联合干预抑制氧化应激、自噬相关蛋白Beclin-1和LC3B-Ⅱ/LC3B-I以及BAX和Caspase-3表达并促进Bcl-2表达. Objective To investigate the effect of electroacupuncture combined with running wheel training on nerve cell autophagy,apoptosis and neurogenesis in the middle cerebral artery embolism(MCAO)rat model.Methods A total of 30 SI)rats were randomly divided into MCAO model group(n=10),combined intervention group(n=10)and sham operation group(n=10).The neurological score was recorded and the infarct volume was measured.The expression levels,(the autophagy effector Berlin 1,the membranous mirmtiiluiⅡ-associated protein 1 A/l B-light chain 3(LC3)(LC3B-Ⅱ)/cytoplasmic LC3(LC3B-Ⅰ),the apoptosis related proteins[B-cell lymphoma-2(Bcl-2),the BCL2-associated X protein(BAX),and caspase-3],p-AMPK,and p-mTOR were detected.Vhr apoptosis of nerve cells was detected.The superoxide dismutase(SOD)and catalase(CAT)activities,and the malondialdehvde(MDA)content were assayed.Results In the MCAO model group,the number of Tunel positive cells,the nervous system score,the cerebral infarction volume,the expression levels of BAX,caspase-3,Bedin-1,LC3B-Ⅱ/LC3B-Ⅰ,p-AMPK,and the MDA content were increased,while the SOD activity,the CAT activity,and the expression levels of p-mTOR and Bcl-2 were decreased(P<0.05).In the combined intervention group,ihe number of Tunel positive cells,the nervous system score,the cerebral infarction volume,the expression levels of BAX,caspase-3,Bedin-1,LC3B-Ⅱ/LC3B-Ⅰ,p-AMPK,and the MDA content were decreased,while the SOD activity,the CAT activity,and the expression levels of Bcl-2 and p-mTOR expression were increase(P<0.05).Conclusion The intervention treatment of electroacupuncture combined with running wheel training inhibits the oxidative stress,suppresses the expression of Beclin-1,LC3B-Ⅱ/LC3B-Ⅰ,BAX,caspase-3,and enhances the expression of Bcl-2 in nerve cells.
