罗布麻AP2/ERF转录因子全基因组鉴定及生物信息学分析

摘要 AP2/ERF类转录因子存在于植物中并对植物生长、发育和逆境响应有重要的调控作用。为研究罗布麻(Apocynum venetum)中AP2/ERF转录因子家族成员性质,本研究基于罗布麻的全基因组数据,结合生物信息学研究方法,对罗布麻中AP2/ERF转录因子进行鉴定和分析。在罗布麻中共鉴定102个AP2/ERF转录因子,结合保守结构域特征和拟南芥(Arabidopsis thaliana)家族成员序列比对,可以分为4个亚家族:AP2、RAV、ERF和DREB。根据生物信息学的分析,该家族的转录因子都是亲水性蛋白质,富含酸性氨基酸,并且大多数成员是非分泌蛋白质。根据染色体定位,100个基因不均匀分布在罗布麻的11条染色体上,多条染色体上的AP2/ERF基因存在串联复制现象。芯片表达分析表明102个成员在植物的生长发育和逆境反应中起着重要作用,可以被低温、高温、光照、遮光、紫外线和脱落酸(ABA)处理诱导,并可以在根、茎、叶、花、果实和种子中表达。同时结合蛋白互作分析及启动子分析推测该基因家族成员在罗布麻的生长发育及对低温、干旱及盐胁迫等多种逆境的生理反应过程中具有重要作用。本研究将为罗布麻AP2/ERF基因家族后续的功能验证奠定基础,同时也在其他物种的AP2/ERF转录因子家族的抗逆性功能研究中具有重要意义。 AP2/ERF transcription factors exist in plants and have important regulatory effects on growth,development and response to stress.In order to study the properties of AP2/ERF transcription factor family members in Apocynum venetum,the AP2/ERF transcription factors in A.venetum were identified and analyzed based on the whole genome data and bioinformatics methods.A total of 102 AP2/ERF transcription factors were identified in A.venetum.Combined with conserved domain characteristics and Arabidopsis thaliana family member sequence alignment,they can be divided into 4 subfamilies:AP2,RAV,ERF and DREB.According to the analysis of bioinformatics,the members of this family are hydrophilic proteins,rich in acidic amino acids,and most of the members are non-secreted proteins.According to chromosome location,100 genes are unevenly distributed on 11 chromosomes of Apocynum venetum,and AP2/ERF genes have tandem duplication on multiple chromosomes.Microarray expression analysis showed that 102 members play an important role in plant growth and development and stress response.The AP2/ERF transcription factors can be induced by low temperature,high temperature,light,shading,ultraviolet and Abscisic acid(ABA)treatment,and can be expressed in roots,stems,leaves,flowers,fruits and seeds.At the same time,according to functional connection network analysis and promoter analysis results of 102 AP2/ERF transcription factors of A.venetum,it is speculated that members of this gene family play an important role in the growth and development of A.venetum and the physiological response to various stresses such as low temperature,drought and salt stress.This study will lay the foundation for the subsequent function verification of the AP2/ERF gene family of A.venetum,and it will also have important significance for the research on the stress resistance function of the AP2/ERF transcription factor family in other species.
