代谢综合征大鼠主动脉Apelin及其受体的表达及意义

The Expression and Significance of Apelin and Its Receptor on the Aorta in Metabolic Syndrome Model Rats

目的观察代谢综合征(MS)大鼠主动脉Apelin及其受体(APJ)的表达变化。方法16只雄性SD大鼠按随机数字表法均分为对照组和MS组,每组8只。MS组采用高果糖饲料(60%)喂养,对照组采用常规饲料喂养,喂养周期为10周。采用Masson染色法检测血管胶原沉积情况,酶联免疫吸附法检测大鼠血清和主动脉组织Apelin含量,免疫组化法检测大鼠主动脉组织APJ的表达。结果高果糖饮食可成功诱导高血压、高三酰甘油(TG)血症和高血糖等特征的代谢综合征模型,MS组大鼠主动脉胶原沉积明显,血清Apelin浓度和主动脉组织Apelin/APJ表达较对照组明显下调(P<0.05)。结论MS大鼠主动脉发生重构,Apelin/APJ表达下调。提示Apelin可能为新的血管保护因子。

Objective To investigate the expression of Apelin and its receptor(APJ)on the aorta of rats with metabolic syndrome(MS).Methods Sixteen SD rats were randomly divided into control group and experimental group,with 8 rats in each group.The experimental group(MS group)was fed with high fructose diet(60%),and the control group was fed with conventional diet.The feeding cycle was 10 weeks.The expression of collagen was determined by Masson staining,Apelin contents in serum and aorta were determined by elisa,and the expressions of APJ were determined by immunohistochemistry.Results The MS model of rats were induced by high fructose diet successfully,with hypertension,hypertriglyceridemia and hyperglycemia.Masson staining showed that there were collagen deposited in aortic wall of MS rats.The contents of Apelin in serum and the expressions of Apelin/APJ in aorta were obviously down-regulated in MS group(P<0.05).Conclusion Vascular remodeling occurred in MS rats,and the Apelin and APJ were significantly down-regulated.It is suggested that Apelin may be a new vascular protective factor.
